| Objective: The incidence of gastric cardia adenocarcinoma(GCA)keep increasing in resent years.However,the exact mechanisms of GCA still remains unclear for the moment.Therefore,further exploring the genes that related to development and progression of GCA will be considered useful.Sprouty2 was one member of Sprouty family.Sprouty2,a known regulator of receptor tyrosine kinases(RTK),as one such regulator.Sprouty2 protein was related to tumors significantly.So it becomes a hot topic of researches at present.In this paper,we investigated the expression of Sprouty2 mRNA and protein in GCA tumor tissues and corresponding tissues,and we tried to analyze the relationship of Sprouty2 expression and clinical data.Methods:1 Reverse transcription polymerase chain reaction(RT-PCR)method was used to detect the mRNA level of Sprouty2 gene in GCA tumor tissues and corresponding normal tissues.2 Immunohistochemistry method was used to examine the protein expression of Sprouty2 in GCA tumor tissues and corresponding normal tissues.3 SPSS13.0 was applied to analyze the results of experiment.Results:1 The mRNA expression of Sprouty2:Sprouty2 mRNA expression in tumor tissues(0.20±0.07)was signifycantly lower than that in corresponding normal tissues(0.66±0.17)(t=-20.868,P <0.05).The mRNA expression of Sprouty2 was not associated with gender and age(P>0.05).The mRNA expression of Sprouty2 gene in high differentiation group(0.24±0.06)was much higher than that in poor differentiation group(0.18±0.06)(t=3.413,P<0.05).The mRNA expression of Sprouty2 gene in non lymph node metastasis group(0.24±0.07)was significantly higher than that in lymph node metastasis group(0.18±0.06)(t=-4.184,P<0.05).The mRNA expression of Sprouty2 gene inⅠ+Ⅱ(0.23±0.08)was significantly higher than that in Ⅲ+Ⅳ stage(0.18±0.06)(t=2.581,P<0.05).2 The protein expression of Sprouty2:The protein expression of Sprouty2 in tumor specimens(31.9%,22/69)was significantly lower than that in corresponding normal tissues(87.0%,60/69)((χ~2=43.395,P<0.05).The protein expression of Sprouty2 was not associated with gender and age(P >0.05).The protein expression of Sprouty2 gene in high differentiation group 52.2%(12/23)was much higher than that in poor differentiation group 21.7%(10/46)((χ~2=6.54,P <0.05).The protein expression of Sprouty2 gene in non lymph node metastasis group 50%(15/30)was significantly higher than that in lymph node metastasis group17.9%(7/39)((χ~2=8.021,P <0.05).The protein expression of Sprouty2 gene inⅠ+Ⅱstage 51.7%(15/29)was significantly higher than that in Ⅲ+Ⅳ stage 17.5%(7/40)((χ~2=12.566,P <0.05).3 The relationship between mRNA expression of Sprouty2 and its expression of protein:The mRNA expression of Sprouty2 in tumor tissues with positive protein expression of the gene(0.26±0.06)was significantly higher than that in tumor tisssues with negative protein expression of the gene(0.18±0.06)(P <0.05).Conclusions:1 Decreased mRNA and protein expression of Sprouty2 was observed in GCA and was associated with development of GCA.2 Decreased mRNA and protein expression of Sprouty2 in GCA was associated with pathological differentiation,lymph node metastasis,and TNM.It suggested that Sprouty2 maybe associated with metastasis and infiltration of GCA. |
