A compound Chinese medicine called Bushen Huoxue prescription contains Rehmannia glutinosa Libosch., Salvia miltiorrhiza Bge., Panax ginseng C.A.Mey. and Pueraria thomsonii Benth.. Modern pharmacological studies show that Bushen Huoxue prescription possesses multiple mechanisms and pathways in the prevention and treatment of DR, including increasing in glutamine synthetase activity, improving glutamate metabolism in retinal Miiller cells, reducing excitotoxicity of glutamate metabolism and so on, which can enhance the activity of retinal Muller cells and strengthen cell membrane stability. However, few studies on the chemical material basis of Bushen Huoxue prescription have been reported. Therefore, in this paper, we research the chemical material basis of the compounds as the fallowing aspects:To begin with, the relationship between fingerprints of this prescription and its relevant herbs was researched. The fingerprints of Bushen Huoxue prescription and its relevant herbs were established by HPLC-DAD-ELSD. The retention time and UV spectral information of chromatographic peaks were taken as indices. Twenty-three common peaks were demarcated in the HPLC-DAD fingerprint and twelve common peaks were demarcated in the HPLC-ELSD fingerprint of Bushen Huoxue prescription. Among them, ten peaks came from S. miltiorrhiza, nine peaks came from P. thomsonii, nine peaks came from P. ginseng, and R. glutinosa did not make a great contribution to the common peaks. In addition, one peak came from S. Miltiorrhiza and P. thomsonii, and another six peaks could not be identified. In the next place, there was designed to established methods for determining the content of danshensu, puerarin, daidzin, salvianolic acid B and ginsenosides in Bushen Huoxue prescription. Ten batches of samples were determined. The results showed that the content of five ingredients from different samples existed discrepancy. It may relate with the production place and storage time of herbs. Then, serum pharmacochemistry of Bushen Huoxue prescription was preliminarily studied, and seven prototype constituents to blood were detected. The might be the active components of prescription. In the end, the influence of different combinations of Bushen Huoxue prescription on the content of acteoside, puerarin, daidzin and daidzein. The content of the five ingredients of different combinations show a dynamic course. However, because the traditional chinese medicine contains many complex components, the law of compatibility is not summarized. The researches above mentioned lay a foundation for studies of quality control and pharmacological material basis of Bushen Huoxue prescription, which also provide an experimental basis for further study of pharmacological mechanism in the prevention and treatment of DR.
