Objective:To investigate the influence of Ziyin Huoxue Jiedu Chinese Herbs on the Vascular Endothelial Cell's damage and the change of adhesion molecules and blood coagulation related factors expression induced by Advanced Glycation End-products,further exploring the protection.Methods:Using human umbilical vein endothelial cell as the subjects,HUVEC cell was damaged induced by AGEs-HSA according to the final concentration(100mg/L).The model group and drug group,respectively,was continue training 12 h,24 h,48 h.To observe each cell morphological changes Under a microscope,testing each adhesion molecule VCAM-1,ICAM-1 expression by western-blot,and detecting the expression tissue factor(TF)and the blood clots regulating element(TM)mRNA expression by rt-pcr method.Result :Compared with the blank group of cells,Model group cell was abnormal proliferation,cell number increased obviously,most of the cell deformation shrinkage,cell membrane hazy outline,a rough sample are observed in the cell changes,VCAM1,ICAM-1,TF expression significantly raised,TM expression reduced.And the most significant effect was after 24h(P<0.01).The same point in time,Compared with model group,Dosage group's circle indented form cell decrease,it can significantly reduce VCAM-1,ICAM-1,TF expression,raise TM expression as time longer effect more apparent.(P <0.01)Conclusions : Ziyin Huoxue Jiedu Chinese Herbs can inhibit the damage on vascular endothelial cell induced by AGEs,reduce the expression of adhesion molecules,reduce the expression tissue factor mRNA transcription,raise blood clots regulate m RNA transcription.
