| 【Objective】1.Understanding of coal tar pitch on occupational exposure to workers health damage.2.Explore the exposure markers and and genetic damage of coal tar pitch.3.Research on gene polymorphism of contact workers urine metabolism of coal tar pitch and the genetic damage repair.【Methods】1.Investigated coal tar pitc h occupational exposure workers,collected worker health information peripheral blood,urine and other biological samples.2.HPLC/MC were used to detect metabolites concentration of occupational exposure workers in urine;multiple PCR technology were used to detect whole blood metabolic gene polymorphisms.3.Single cell gel electrophoresis were used to detect genetic damage.Endpoint method technique to detect whole blood repair gene polymorphism.4.Use spss17.0 for statistical analysis.5.Study of coal tar pitch with the method of comprehensive analysis o f contact workers health damage and to explore its biomarkers.【Results】1.Field investigation resultsThe type of work contact with coal tar pitch in deep processing of coal tar pitch(production enterprises)company are tar work,asphalter,tank operator,Electrical fitter,Analysis work.The main workshop of Coal tar pitch using company(carbon production enterprises)are shaping shop and roasting workshop.The type of work.are Fabric work,Pack out work,overhead crane operator overhead crane operator.2.Air samples field test resultsThe highest average exposure concentration of TWA is 0.63mg/m3,The minimum average exposure concentration of TWA is 0.07 mg/m3,the content of coal tar pitch in Air samples with the type of work have significantly correlation(R=0.83,P<0.05).3.Healthy physical examination resultsContact with coal tar pitch group and control group in age,length of service whether smoking and drinking differences have no statistical significance(P > 0.05).Anomaly detection rate of the skin is 10.61%.FEV1.0 % in Contact group is higher than the control group,the difference was statistically significant(P<0.05).Contact group of systolic and diastolic blood pressure is less than the control group,the difference was statistically significant(P<0.05).Contact group of WBC,MEUT,GRA,MCHC is significantly higher than the control group.Contact group of LYN%,RBC,HGB,RDW-SD is significantly lower than the control group(P<0.05).4.Laboratory test results4.1PAHs metabolites in urine test resultsContent in contact group of 2-OH-N,1-OH-P,1-OH-N is significantly higher than the control group(P<0.05).Urine metabolites content of PAHs have correlation with coal tar pitch in workplace air,correlation-coefficient of 2-OH-N,1-OH-P,1-OH-N is 0.40,0.21,0.40(P<0.05).Smoking on workers urine co ntent of 2-OH-P,1-OH-P,1-OH-N were statistically significant(P<0.05).Content of 2-OH-P,1-OH-P.in Molding workshop workers urine is highest,Preparation workshop have the highest content in1-OH-N(P<0.05).Metabolites in urine significantly higher during work(P<0.05).4.2Analysis of genetic damage in peripheral blood cellsContact group tail DNA %and O live tail is significantly higher than the control group(P<0.05).The tail DN A % in Smoking group is higher than not smoking group,the difference was statistically significant(P<0.05).The tail DN A %and O live tail in Molding workshop workers get the highest levels.Difference was statistically significant(P<0.05).Metabolites in urine have relevance with the tail DNA % and Olive tail,the difference was statistically significant(P<0.05).4.3Genetic polymorphisms of the detection2-OH-N,1-OH-P,1-OH-N were significantly different in different GSTT1 genotype and GSTM1 genotype(P<0.05),the interaction of GSTT1、GSTM1has significantly difference in 2-OH-N,1-OH-P(P<0.05).The interaction of XRCC194 and XRCC280 has significantly difference in tail DNA %and O live tail(P<0.05).Length of service,smoking of the effect of DNA %and O live tail moment difference was statistically significant(P<0.05).【Conclusions】1.Occupational exposure to coal tar pitch can cause skin damage,such as Skin prick,skin pigmentation,skin itch,comedones,and even headache systemic symptom.2.Occupational exposure to coal tar pitch can cause damage to the lung function and blood pressure,blood routine examination.3.Occupational exposure to coal tar pitch,and smoking affect the content of 2-OH-N,1-OH-P,1-OH-N.2-OH-N,1-OH-P,1-OH-N can be used as the exposure indicator contact with the content of coal tar pitch.4.Occupational exposure to coal tar pitch can cause genetic damage of the workers,2-OH-N,1-OH-P,1-OH-Ncan indirectly reflect the degree of genetic damage.5.GSTT1,GSTM1 gene polymorphism,the interaction of GSTT1 and GSTM1 Impact on metabolism of the coal tar pitch in body.the interaction of XRCC194 and XRCC280 effect on the body of genetic damage repair. |
