Objective: To determine whether protein kinase 1,protein kinase 4 can be a new tumor marker or prognostic factor for non-small cell lung cancer and to assess this possibility,we investigated and used experimental methods to determine patients with non-small cell carcinoma Lung cancer tissue and adjacent normal lung tissue PAK1,PAK4 expression,combined with statistical analysis of non-small cell lung cancer patients with clinical and pathological features,prognosis and the two organizations PAK1,PAK4 expression between the association.Methods:This study included 186 patients with non-small cell lung cancer who underwent surgical treatment between January 2012 and December 2012 in Shandong Provincial Tumor Hospital.All patients had complete clinical and pathological data.All patients undergoing surgery did not receive any other treatment,such as radiotherapy,systemic chemotherapy or immunotherapy,and the specimens removed during the procedure must be diagnosed by at least two pathologists to determine the type of pathology.All cases were traced back to December 31,2016 or the time of death from the beginning of the treatment to the last follow-up of complete death(for any reason)or complete data,followed by 79 cases.(P <0.05).There were no significant differences in real-time quantitative reverse transcription-polymerase chain reaction(RT-PCR),which was used to detect RNA from lung cancer tissues and adjacent tissues(5cm or more,PCR)to detect the expression of PAK1 and PAK4 genes.At the same time,the paraffin sections of these non-small cell lung cancer tissues were stained with immunohistochemical method to distinguish the expression levels of protein kinase-1 and protein kinase IV.At the same time,careful follow-up of pathology,and finally analysis of whether there is a relationship between the two,used to prove whether these two protein kinase can be used as non-small cell lung cancer prognosis indicators.Results: The relative expression of PAK1 m RNA in non-small cell lung cancer tissues and adjacent normal tissues was 0.454 ± 0.37 and 0.30 ± 0.37,respectively.The expression of PAK1 m RNA in patients with non-small cell lung cancer was 0.454 ± 0.37 and 0.30 ± 0.37,respectively.The content was significantly higher than that of normal tissues(P <0.01).The relative expression of PAK4 m RNA in non-small cell lung adenocarcinoma tissues and adjacent normal tissues was 0.70 ± 2.31 and 0.41 ± 1.01,respectively.The content of PAK4 m RNA in tumor tissue was higher than that in normal tissues,and there was no significant difference(P> 0.05).Immunohistochemical results showed that PAK1 staining in non-small cell lung cancer tissue was stronger than that in normal tissues.PAK1 protein expression was positively correlated with m RNA.PAK1 protein expression and gender,pathological type and so there is a significant correlation.The expression of PAK4 protein was not correlated with sex,age,pathological type,differentiation degree,lymph node metastasis and staging.The high expression rate of PAK1 in non-small cell carcinoma was 89.7%,and the high expression rate of PAK1 in non-small cell adenocarcinoma was 78.3%(P <0.05).However,in patients with age,differentiation degree,lymph node Metastasis,staging and other aspects of PAK1 expression no significant difference(P> 0.05).Thus,the expression rate of PAK1 in female adenocarcinoma was significantly higher than that in female adenocarcinoma(P <0.05).According to the protein expression score of P-21 activating kinase 1,4 in the tumor tissue,the patients were divided into low expression group and high expression group.The overall survival time of PAK1 overexpression group was 34.60 ± 2.562 months,and the mean survival time of PAK1 low expression group was 39.576 ± 2.331 months,and the difference was not statistically significant(P = 0.237)(Figure 3).(P = 0.003)(P <0.05),but there was no statistical significance in the other stages of stage ?(P <0.05),but there was no significant difference between the two groups(P <0.05)The The mean survival time of PAK4 high expression group was 37.60 ± 3.801 months,the mean survival time of PAK4 low expression group was 36.214 ± 1.012 months,the difference was not statistically significant(P = 0.154)(Figure 5)The survival analysis showed that there was no significant difference in PAK4 expression between patients with stage III non-small cell lung cancer(P = 0.237)CONCLUSION: PAK1 is overexpressed in non-small cell carcinoma tissues.PAK1 protein expression is significantly associated with gender and pathological type,which is highly expressed in PAK1 expression in female adenocarcinoma,and the difference is significant(P <0.05).The up-regulation of PAK1 in non-small cell lung cancer indicates that PAK1 may be involved in the carcinogenesis and development of women with adenocarcinoma of non-small cell lung cancer.PAK1 is expected to be one of the predictors of poor prognostic factors in non-small cell lung cancer patients.PAK4 has not been found as a non-small cell lung cancer adverse prognostic indicators.
