Establishment Of Rats Hepatocellular Carcinoma Model And Effect Of ICG-001 On Survivin In Rats Induced Hepatocellular Carcinoma

Objective: To establish hepatocellular carcinoma(HCC)model in SD rats and to investigate the effect of β-catenin inhibitor ICG-001 on expression of Survivin in hepatocellular carcinoma of SD rats by using ICG-001 in vivo.Methods : SD rats were fed with diethylnitrosamine(DEN),and then the rats which have tumor were divided into control group,DMSO group and administration group.ICG-001 was used to inhibit the expression of β-catenin in administration group by intraperitoneal injection.Western blot was used to detect the effect of ICG-001 on the protein expression of Survivin.The RTPCR was used to detect the effect of ICG-001 on the mRNA of Survivin.The protein and mRNA expression levels of Survivin were detected by ImageJ software and gel imaging analysis system.The gray values of bands were measured and transformed into data,and then using SPSS 18.0 to analysis.Results: The success rate of establishing the HCC model by DEN was83.3%.Western blot analysis showed that the protein expression of Survivin decreased after administration of ICG-001:the expression of Survivin inadministration group was significantly lower than that in control group,and the difference was statistically significant(p = 0.001 <0.05),while between the control group and the DMSO group the difference was not statistically significant,p = 0.428> 0.05.The RT-PCR results showed that the mRNA expression of Survivin was decreased after administration of ICG-001:the mRNA expression of Survivin in administration group was significantly lower than that in control group,and the difference was statistically significant(p=0.036< 0.05),while between the control group and the DMSO group the difference was not statistically significant,p=0.456>0.05.Conclusions: The success rate of establishing the HCC model by DEN was 83.3%.β-catenin inhibitor ICG-001 significantly inhibit the protein expression and mRNA expression of Survivin.
