Shuang Huang Tiao Zhi prescription is a Chinese herbal medicine compound of Turmeric,Rhizome Coptidis and Pueraria for reducing blood lipid on the basis of previous experimental study and clinical prescription.Studies have shown that curcumin,berberine and puerarincan are the effective ingredient of this prescription.In this study,we used animal and cell models in vitro to analyze the effect of Shuang Huang Tiao Zhi prescription to the lipid levels and lipid metabolism-related enzymes explored the lipid-lowering effect and possible mechanism of Shuang Huang Tiao Zhi prescription and provided certain basis for the drug development and clinical application.In the present study,we used tail intravenous injection of Trtion-WR 1339 to establish the endogenous hyperlipidemic mice models,intraperitoneal injection of egg yolk emulsion to establish the exogenous hyperlipidemic mice model,then the lipid-lowering effect of Shuang Huang Tiao Zhi prescription on the pathological models were studied.In vitro,the influence of Shuang Huang Tiao Zhi prescription to the contents of cholesterol(CHO),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein receptor(LDL-R),3-hydroxy-3-methylglutaryl-CoA(HMG-CoA)reductase,cholesterol7α-hydroxylase(CYP7A)and lipoprotein lipase(LPL)in HepG2 cells were studied.HepG2 cells were induced by oleic acid(OA)and treated different concentration of curcumin,berberine and puerarincan,which are effective components of Shuang Huang Tiao Zhi prescription.Results:(1)Shuang Huang Tiao Zhi prescription with a concentration of 22.5 g/kg can significantly decrease serum CHO and TG level in endogenous hyperlipidemia mice model induced by Trtion-WR 1339.(2)Shuang Huang Tiao Zhi prescription with a concentration of 22.5 g/kg can significantly decrease serum CHO and TG level in exogenous hyperlipidemia mice model induced by egg yolk emulsion.(3)Curcumin with a concentration of 10 μg/mL can significantly increase HDL-C,LDL-R,LPL content and decrease TG,HMG-CoA reductase content in HepG2 cells induced by OA.(4)Berberine with a concentration of 40 μg/mL can significantly increase CHO,HDL-C,LDL-R,CYP7A1 content in HepG2 cells induced by OA.(5)Puerarincan with a concentration of 300 μg/m L can significantly increase HDL-C,LDL-R,CYP7A1,LPL content and decrease CHO,TG,HMG-CoA reductase content in HepG2 cells induced by OA.Conclusion: Shuang Huang Tiao Zhi prescription can significantly decrease blood lipids of hyperlipidemia mice caused by endogenous and exogenous factor.Curcumin,berberine and puerarincan which are the effective ingredients of Shuang Huang Tiao Zhi prescription could reduce blood lipid by inhibiting endogenous CHO synthesis,promoting CHO transformation,promoting the reverse cholesterol transport(RCT)pathway,promoting the LDL-R pathway and degradation of TG.The pharmacodynamic basis and molecular mechanism of lipid-lowering effect of Shuang Huang Tiao Zhi prescription are further clarified.
