Human gingival fibroblasts were used as the negative control cells and human calvarial osteoblasts were used as the target cells for whole cell biopanning from a12 mer peptide phage display library to obtain the peptide that specifically bind to osteoblasts.Its amino acid sequence is VLAVPWSEPGYL(valine-leucine-alanine-valine-proline-tryptophan-serine-glutamic acid-proline-glycine-winding glycine leucine).The experiment based on tissue engineering using chitosan and alginate as the scaffold loaded with the peptide that specifically bind to osteoblasts as signal factor implanted into the rabbit skull bone defect,to evaluate the effetct of bone regeneration of the peptide.To solve the problems of bone defects of cranial、maxillofacial and oral implantation,this research provide a new idea and treatment.Specific content is as follows:The first part:the preparation of alginate/chitosan/peptide compound gelUnder aseptic conditions,mix alginate/the peptide suspension with chitosan solution,keep them in solution condition and distribute evenly the peptides through mixing reaction.Regulate the final concentration of the peptide to 160μg/ml and prepare sodium/chitosan/the peptide compound gel in a certain temperature.Prepare compound gel without the peptide used as the control group in the experiment.The second part: the preparation of critical size defects of rabbit skull12 healthy new zealand white rabbits were used to prepare bilateral parietal critical size bone defects of 15 mm diameter.SA/CS/Peptide Gel was implanted into the experimental group,SA/CS Gel was implanted into the control group and the empty group without scaffold or drug,all defects randomly divided into groups.The bone regeneration ability was investigated by CBCT,High-frequency molybdenum target X-ray,Hematoxylin and eosin staining(H&E)at 8 weeks after surgery.According to imaging and histomorphology,the experimental group showed the highest bone regeneration than the control group and the empty group.The difference have statistical significance(P < 0.05).Conclusion The peptide that specifically bind to osteoblasts can enhance the bone regeneration of rabbit calvarial defect.
