Background:Scar is one of the common clinical disease,it can be divided into pathological scar and physiological scar.Scars often form after trauma,the apropriate scar formation is a physiological and self-defense performance,but the scar’s over-proliferation is a pathological performance.Pathological scar can be divided into hypertrophic scars and keloids,which often lead to adverse effects of patient’s appearance and function.Pathological scar will also cause psychological stress and burden in modern society.With the economic development and the living standards improvement,peoples’ requirements of life quality and beauty are getting higher,so the etiology exploration,new diagnosis and treatment technologies for Pathological scar are more needed.Two major characteristics of pathological scar are fibroblasts’ over-proliferation and tumor-like characteristics.The present study shows that fibroblasts are the main cells of wound healing,which are activated,proliferated and synthesized in the process of wound healing,and their over-proliferation is a major feature of pathological scar.Pathological scar have similar biological characteristics with the,such as the etiology,pathology,clinical manifestations,diagnosis,treatment and other aspects,that is.On the one hand,the tumor-like characteristics of pathological scar can due to better overview of the biological characteristics for pathological scar,on the other hand,it can reveal the complexity and the difficulty of pathological scar prevention,so the pathogenesis research of pathological scar can learn from the research ideas and achievements of the tumors.In 2005,some scholars found that Hippo signaling pathway in the fruit fly can transmit cellular signals from the cytoplasm to the nucleus for the first time,it regulate the expression of downstream genes.In addition,it also regulates cell growth,proliferation and apoptosis,and participating in the regulation of organ size and tissue regeneration.Hippo signaling pathway have an important role in the development and organization of organs,it also have a direct relationship with the occurrence and development of tumors.In conclusion,we hypothesized that the Hippo signaling pathway plays a role in the pathogenesis of pathological scar formation.The expression of YAP/TAZ plays a key role in the Hippo signaling pathway.Therefore,this study intends to describe the role of the Hippo signaling pathway in pathology scar.Purpose:To study the expression of YAP/TAZ in human dermal fibroblasts and hypertrophic scar fibroblasts,and to establish the model of rabbit ear hypertrophic scar,and to detect the expression of YAP/TAZ in hypertrophic scar of rabbit ear.Method:(1)Flow cytometry was used to detect the apoptosis of human dermal fibroblasts and hypertrophic scar fibroblasts.Western Blot,RT-PCR and Q-PCR were used to detect the YAP/TAZ expression of Hippo signaling pathway in human dermal fibroblasts and hypertrophic scar fibroblasts.(2)Rabbit ear scar model was established:use the ring drill to do four 6mm round full-thickness skin wounds in each rabbit ear.The wound interval is 2cm or more.In this process we remove the perichondrium,which can promote hypertrophic scar formation.We use HE staining to prove hypertrophic scar formation and the expression of YAP/TAZ was detected by Western Blot.Result:(1)Hypertrophic scar fibroblasts apoptosis is weaker than that of human dermal fibroblasts.YAP/TAZ expression in hypertrophic scar fibroblasts is higher than that of human dermal fibroblasts.(2)Rabbit ear hypertrophic scar forms after injury at about 21 days,HE staining showes collagen hyperplasia andfibroblast proliferation.Conclusion:YAP/TAZ,the key molecules of the Hippo signaling pathway plays a role in the pathogenesis of pathological scars.
