Ferulic acid is an important component of Ligusticum chuanxiong Hort. Many scholars have observed that Sodium ferulate has therapeutic effects on migraine. In this research, we investigate the influence of ferulic acid on plasma concentration of calcitonin gene-related peptide, substance P, and neuropeptide Y in jugular vein in rats after electrical stimulation of the trigeminal ganglion (TG).MethodA total of 48 SD male rats were randomly divided into 6 groups, which were Sumatriptan group 30mg/kg)、high-dose Sodium ferulate group (100mg/kg)、Middle-dose Sodium ferulate group (50mg/kg)、low-dose Sodium ferulate group(25mg/kg)、Sham operation group (1 mL saline)、Saline group (1 mL saline)。The Sumatriptan group、each group of Sodium ferulate and Saline group were prepared model while sham operation group received sham operation after 1ml blood was taken from the jugular vein. 1ml blood was taken from the jugular vein again after electrical stimulation . Then mice in the Sham operation group and Saline intraperitoneally injected with normal saline, mice in other group were intraperitoneally injected respectively with testing medicine. 1ml blood was taken from the jugular vein after 30 minutes. The concentration of CGRP, SP, NPY were detected by ELISA assay.Result:①Comparison of concentration changes of plasma SP at 30 minutes after treatment:The SP concentration of all the sodium ferulate group were decreased compared with the normal saline group and the decreased ratio were significantly different; The decreased ratio had no significant difference between Sumatriptan group and High-dose Sodium ferulate group and Middle-dose Sodium ferulate group.②Comparison of concentration changes of plasma CGRP at 30 minutes after treatment:The CGRP concentration of all the sodium ferulate group were decreased compared with the normal saline group and the decreased ratio were significantly different; The decreased ratio had no significant difference between Sumatriptan group and High-dose Sodium ferulate group and Middle-dose Sodium ferulate group.③Comparison of concentration changes of plasma NPY at 30 minutes after treatment:The average NPY concentration of the saline group and sodium ferulate groups all showed an upward trend and there were no significantly different in the rising ratio.There was a statistically significant difference in the rising rate between low-dose Sodium ferulate group and Sumatriptan group.Conclusion:The effect of Sodium ferulate in migraine treatment may due to dose-dependent inhibition of release of SP and CGRP. | | Keywords/Search Tags: | migraine, Sodium ferulate, Ligusticum chuanxiong, CGRP, SP, NPY | PDF Full Text Request | Related items |
