| Research BackgroundCervical cancer is one of the largest one of malignant tumors harm to women’s health,and the trend to grow,and there are increasingly younger trend.Occurs in cervical squamous columnar epithelium,more for solid tumors,spread development directly.The current treatment is similar to other malignant tumors,with surgery is given priority to,complementary with put,chemotherapy,side effect is obvious.Need a new both can improve the quality of life of patients and treatment can improve the prognosis.Human myoblasts gene therapy(HMGT)is an advanced method currently which has been confirmed more effective in treatment of muscle atrophy and diabetes and cardiomyopathy.The study about HMGT showed that myoblasts could enhance the contraction and relaxation of the heart;the myoblasts transplantation technology applied to treat diabetes achieved good results on animal experiments;in the treatment of malignant tumors,by injecting human myoblasts into mouse models,studies had shown that it could inhibit the growth and metastasis of prostate cancer solid tumors.Research ObjectiveTo establish Preliminary research on the feasibility of the muscle cells in cervical cancer treatment.Establish a nude mice model of cervical cancer,injecting muscle cell after treatment,according to a series of indicators,to a preliminary assessment of the effect muscle mother cells for the treatment of cervical cancer.To explore a new method for cervical cancer treatment.As far as possible improve the patients quality of life,reduce the pain of side effects.Research MethodsNude mice injected with cervical cancer cells after the success of the building,45 successful model for 3 groups at random,as the muscle cell group,saline group,cisplatin group,each group of 15 only,muscle cell tumors had injection respectively,intraperitoneal injection of cisplatin and physiological saline.Then measured the size of tumor,observing the growth of nude mice.Observed after 4 weeks,remove tumors,and using HE staining to observe the change of tumor cell morphology,immunohistochemistry and western testing the amount of protein expression in tumor cells ResultsExperimental muscle before mother cell group,cisplatin group and saline group of nude mice weight and tumor size(16.05 + 0.18 g),respectively(36.71 + 0.52)mm3,g(16.09 + 0.15),(36.77-0.68)mm3,g(16.08 + 0.13),(36.53-0.85)mm3,after the experiment 3 groups respectively corresponding g(16.35 + 0.26),(20.14-1.88)mm3,g(16.09 + 018),(55.42-3.35)mm3,g(15.72 + 0.19),(94.97-4.68)mm3;HE dyed saline group is a large number of typical pleomorphism of tumor cells,tumor cells visible muscle cell group was obviously a lot of apoptosis necrosis,cisplatin group of visible part of the apoptosis necrosis of tumor cells.Immunohistochemical results showed that P53 positive rate are 20.0%,33.3% and 20.0%respectively;Western Blod results show: P53 protein expression relative to the amount of 0.321 + /-0.054,0.379 + /-0.067 and 0.067 + /-0.064,the comparison group,P< 0.05.The expression of P53,quantity has significant difference between three groups.Research ConclusionExperimental results of muscle group mother cell tumors had smaller was statistically significant,HE staining cervical cancer cells that in muscle cell group was obviously cracking,necrosis,immunohistochemical and western that muscle cell group significantly inhibits the expression of P53 protein,presumably muscle cells can inhibit the growth of tumors,promote the apoptosis of cervical cancer cells,inhibit the expression of P53.All levels from tip of muscle cells in the treatment of cervical cancer has certain feasibility,it is possible that after surgery,and a new treatment after chemotherapy. |
