| Research BackgroundIn recent years,with the continuous exploration of female tumor research,cervical cancer has become one of the most common malignant tumors that threaten women’s health following breast and ovarian cancer.The common site of cervical cancer is the junction of squamous epithelium and columnar epithelium,which is the attracting site of human papillomavirus(hpv)and is a lesion of cervical cancer.In recent years,the implementing of cervical cancer screening and the early detection of cervical cancer and early lesions have significantly reduced the incidence and mortality of cervical cancer,but the trend of cervical cancer has become younger.Currently,the main treatment of cervical cancer is surgery and radiotherapy,chemotherapy-assisted comprehensive treatment and other traditional treatment methods.Whether it is through surgery to remove the lesion,or some patients and patients that are not suitable for e surgery can only be treated by radiotherapy,chemotherapy which still bring some serious damage to the body,spirit,psychology,family and society.In order to alleviate the pain of surgery and some side effects of radiotherapy and chemotherapy.Doctors and researchers have been constantly exploring new treatment methods,hoping to explore new treatment methods through scientific research to better treat patients with better quality of life.The treatment of myoblast transplantation for some diseases is a new advanced-treatment technology in the world and it has the characteristics of easy sampling,high value-added ability in vitro culture and low carcinogenic rate.A large number of experiments have confirmed that myoblast transplant treatment has a better effect on a variety of diseases,such as Duchenne muscular dystrophy,ischemic cardiomyopathy,prostate cancer,type II diabetes,urinary incontinence,and so on.Research ObjectiveThis paper mainly studies the effect of myoblasts on cervical cancer and its mechanism.The results will be used as the basis to evaluate the therapeutic effect of different experimental methods on cervical cancer by establishing human nude mice model,grouping and adopting different treatment methods on molecular level,immunohistochemicalization,western blot and so on.Through the results of this experiment,we can identify whether myoblasts can be a new method for the treatment of cervical cancer,and compare the mental and weight changes among different groups and explores the mechanism of myoblast transplantation in the treatment of cervical cancer.Bring more hope for our patients.Materials and Methods30 nude mice which were purched from Weitong Lihua company and were randomly divided into three groups,10 rats in each group were injected with normal saline(0.2 ml/only),cisclocarboplatin(intraperitoneal injection)arid myoblasts(1 x 107).After each group of nude mice were treated,the tumor volume was measured every one day and the growth of nude mice was observed.After 5 weeks of observation,the tumor was removed from the body after successful intraperitoneal 9injection of chloroform(800 mg/kg).HE staining,immunohistochemistry,Western Blot method to further detect the related changes of tumor cells and related proteins in tumor tissues.Research ResultsAfter successful experimental modeling and grouping,we measured the body weight and tumor size of nude mice in the saline group,cisclocarboplatin group and myoblast group.the body weight of the nude mice in the first three groups was(16.13±016)g,(16.08 ± 021)g and(16.09 ± 0.19)g,respectively.the volume was(109.98± 3.92)mm 3,:(111.84±4.72)mm 3,(112.90 ± 4.53)mm 3.The body weight and tumor volume of three groups of nude mice were monitored by the same method.the weight was(16.61 ± 0.26)g,(16.05 ± 0.22)g and(15.75 ± 0.22)g,respectively.the tumor volume was(165.36±5.04)mm3,(133.42±5.62)mm3 was(89.45±4.87)mm3.HE staining:The results of the experiment showed that there was no significant difference between normal saline group and general malignant tumor.We can see the obvious cell lines,intercellular bridges,distinct heterotypic cells,large tumor cells,increased nuclei and fine tumor A small portion of the intracellular fragmented,disintegrated,and necrotic cells are focal focal necrotic foci in the tissue space due to excessive growth of tumor cells and inadequate nutritional supply.In cis-chloroplatin tissue sections,more obvious tumor cell sequestration,nucleus lysis and sequestration can be observed,and the necrotic foci have a significantly larger trend than in the normal saline group.Immunohistochemical results:The positive rates.of Ki67 in normal saline,cis-chloroamplatin and myoblast group were 70.0%,40.0%,20.0%;Her-2 staining results:saline,cis-chlorocarboplatin,myoblast group positive rate are 50.0%,40.0%,10.0%.Western Blot results showed that the Ki67 protein contents in the three groups were 1.187±0.044,0.764,0.145,0.420 ± 0.108,respectively.The statistical analysis showed that there was a difference in protein content between myoblast and saline group(P<0.05).The Her-2 protein contents of the three groups were 0.694 ± 0.043,0.534±0.056,0.460 and 0.093,respectively.Statistical analysis showed that there was a difference in protein content between myoblast and saline group(P<0.05),there were no difference between the other groups(P values>0.05).ConclusionThe results of the three groups of experiments showed that:1.The body weight of naked mice in myoblastic treatment group increased rapidly and the tumor decreased significantly.2.The results of HE staining showed that a large number of cells were disintegrated and necrotic in the myoblast group than in the other two groups.3.Immunohistochemical and Western blot suggested that the contents of Ki67 proteins in myoblast group were significantly reduced.Immunohistochemical of Her-2 showed no difference between groups,while the Western Blot showed that the content of myoblast protein was significantly lower than that of saline group.So we can see from the above several aspects that myoblasts were more effective in treating cervical cancer. |
