Objective: This study was to investigated the postoperative quality of life and the degree of BRCA1 gene methylation in patients with breast cancer in Xinjiang,to understand the prognosis of patientsand its influencing factors,explored the value of BRCA1 gene methylation in the prognosis of peripheral blood,provided the basis for prognosis evaluation and rehabilitation guidance of breast cancer.Methods: 192 primary breast cancer patients(including 66 Uygur,126Han nationality)hospitalizedin XinjiangTumorHospitalfrom 2011 to 2012 were followed up by face-to-face interview.The questionnaire of FACT-B were filled.The clinical data and peripheral blood sampleswere collected.The methylation of BRCA1 genewas detected by pyrosequencing.The questionnaire was analyzed by descriptive statistics,t test,ANOVA and multiple stepwise regression analysis.The data of biological samples were analyzed by rank test,correlation test and Spearman rank correlation analysis.Results: The average score of life quality in breast cancer patients was 84.33 ± 16.5,physical statuswas 18.01 ± 5.4,social / family statuswas 17.18 ± 4.02;emotional statuswas 12.45 ± 3.08;functional statuswas 16.48 ± 4.52;additional statuswas 20.21 ± 4.16.Univariate regression analysis showed that factors affecting the quality of life score and other status were age,nationality,education level,monthly income,marital,recurrence or not,clinical stage,health care;multiple stepwise regression analysis showed that ethnicity,age,clinical stage,whether recurrence and metastasis and monthly income impacted the quality of life much greater.Compared the methylation rate of BRCA1 gene promoter in the peripheral blood samples after operation showed that BRCA1 gene PM00064862 CpG 1,Cp G2,and CpG3,PM00064876 CpG 1,CpG 4 was statistic significant between with recurrence and metastasis or not recurrence and metastasis;the methylation rate of BRCA1 gene in patients with recurrence and metastasis before operation and after operation was compared: there was no statistic significant in the methylation rate between region PM00064862 and region PM00064876;the methylation rates of BRCA1 gene was compared between the patients without recurrence and metastasisbefore operation and after operation,there was significant difference in the methylation rate of region PM00064862CpG1,Cp G3,CpG4 and PM00064876CpG1,Cp G2(P<0.05).The methylation rate of BRCA1 gene in different pathological types in region PM00064862 CpG1,CpG2,Cp G3,CpG4 and region PM00064876 CpG1 was statistically significant.The methylation rates of BRCA1 gene was compared between ER negative and positive groups,there was statistically significant in the methylation rates of region PM00064862 CpG1,CpG3 was(P<0.05);the methylation rates of BRCA1 gene were compared between PR negative and positive groups,themethylation rates of region PM00064862,CpG1,CpG2,CpG3 was statistically significant(P<0.05);the methylation rates of BRCA1 gene were compared between Her-2 negative and positive groups,themethylation rates of region PM00064862 CpG1,Cp G3 and PM00064876 Cp G1 was statistically significant(P<0.05);the methylation rates of BRCA1 gene were compared between the triple-negative and not triple-negativepatients,there was statistically significant in the methylation rates of regionPM00064862 CpG1 and region PM00064876 Cp G1(P<0.05);the methylation rates of BRCA1 gene PM00064862 CpG1,CpG2,CpG3 was negatively correlated with ER,themethylation rates of BRCA1 gene PM00064862CpG1,CpG2,CpG3,Cp G4 was negatively correlated with PR,the methylation rates of BRCA1 genePM00064876 CpG1,Cp G2,CpG3,CpG4 was positively correlated with HER-2.Conclusion: Nationality,age,clinical stage,recurrence and metastasis,monthly income impacted the quality of life of women after breast cancer surgery in Xinjiang area.The methylation of CpG island in the promoter region of BRCA1 gene in peripheral blood may be the hint for the prognosis of breast cancer.
