ObjectiveThis article was to investigate the meaning of joint detecting p16 and CDO1 genes methylation in plasma in the diagnosis of glioma.Methods Methylation-specific polymerase chain reaction(MSP)were to detect the status of the genes methylation on p16 and CDO1 in the tumor tissues and the corresponding preoperative blood plasma of 49 patients with glioma,cerebral contusions and the corresponding preoperative blood plasma of 10 patients with craniocerebral trauma,and the plasma of 16 people with health check-up,in order to investigate the value of p16 and CDO1 genes methylation in the clinical diagnosis of glioma.ResultsThe p16 and CDO1 genes methylation ratesin glioma tissues were 53.1%(26/49)and 42.9%(21/49),and the corresponding plasma methylation rates were 39.0%(19/49)and 34.7%(17/49).The genes methylation frequency for either p16 or CDO1 or both in tumor tissues and in corresponding plasma were 75.5%(37/49)and 67.3%(33/49).The joint detected sensitivity of glioma by p16 and CDO1 genes in plasma is 67.3%,an obvious improvement compared to the sensitivity detected by a single gene.Meanwhile,the methylation were not detected abnormal in the plasma from the control group of 10 cases of cerebral contusions and 26 cases of plasma,indicating that the detected specificity alone or joint by the p16 and CDO1 methylation were 100%.Univariate analysis found that p16 and/or CDO1 methylation in plasma from the preoperative patients had no significant correlation with age,gender,and tumor grade.Conclusion The joint detecting on the methylation status of some tumor specific genes can enhance the sensitivity of the glioma at the serological diagnosis level,providing a new concept at the clinical diagnosis level in regards to screening and the early diagnosis of glioma.
