Lung cancer,a complex molecular network disease,is one of the most common malignancies tumor around the world at present.The effective treatment of lung cancer now is full resection with appropriate chemotherapy.Therefore,the most important treatment for lung cancer is still looking for biomarkers which facilitate early diagnosis and prognosis.In this paper,we analyze the coding genes and 1ncRNA genes by using differential expression analysis(DEA)and weighted gene co-expression network analysis(WGCNA),respectively.Using the WGCNA method,we constructed a gene co-expression matrix,made cluster analysis to obtain forty seven modules,eight of them are lung adenocarcinoma risk modules.And 27%of these eight module genes are differentially expressed genes.Then,we performed gene ontology(GO)and KEGG functional enrichment analysis for the eight modules and the differentially expressed genes(DEG)found by DEA.We found that WGCNA method can find some biological process which couldn’t enriched by DEA.For example,the WGCNA method precisely gives the direction of collagen molecule regulation in the biological processes associated with the metabolism of collagen biomacromolecules.In eight lung adenocarcinoma risk modules,the DEGs and 1ncRNAs account for 68.9%and 15.4%of the green module genes,respectively,which has the highest proportion of DEG in the eight modules.In blue module,the DEGs and 1ncRNAs account for 8%and 33.3%,respectively,which has the highest proportion of 1ncRNA in the eight modules.The top 50 genes with the highest connectivity in blue module have 16 1ncRNAs.Greenyellow,green and darkred modules have 3 1ncRNAs,respectively.Purple module has 2 1ncRNAs,yellow module has one 1ncRNA.Therefore,1ncRNA may play an important role in the development of lung adenocarcinoma.The hub genes of SPTBN1,SFTPC,FHL1 and RP5-826L7.1 in green module were involved in the development of lung adenocarcinoma,SFTPC has the highest gene significance(GS)value and FHL1 has the highest module membership(MM)value.In the greenyellow module,the hub gene of SAMHD1 plays a role in lung adenocarcinoma through the immune response process.NLRC4 and FCER1G were play a funtion through the immune response process in other diseases.NLRC4 has the highest GS value and FCER1G has the highest MM value.Although there are no reports that these two genes are involved in the development of lung adenocarcinoma,we have reason to believe that the two genes and the hub genes in each module may play a role in the pathogenesis of lung adenocarcinoma.Therefore,the hub gene may serve as an early effective diagnosis and prognosis marker for lung adenocarcinoma.
