| Objective:To investigate the effect of studying protective effect and mechanism of fucoidan derived from Lessonia Nigrescens(LN)on kidneys in diabetic rats.Methods:Streptozotocin(STZ)was used to induce diabetic rats which were divided into six groups of Nor,Fuc-50,Fuc-200,Cap and Mod.The urea nitrogen(UREA),urinary microscale total protein(M-TP),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),serum creatinine(CREA),cholesterol(CHOL),blood glucose(GLU),triglyceride(TRIG),and glycated serum protein(FRUC)were determined using an automatic biochemical analyzer.Elisa was used to quantify the urinary creatinine(u Cr),urinary albumin(u Alb)and renal P-selectin,alpha transforming growth factor(TGF-alpha),interleukin-6(IL-6)and mitogen activated protein kinase/MAP kinase(MAPK).Hematoxylin-eosin staining(HE)was adopted to check the pathological changes in kidney of DN rats.Immunohistochemical was used to quantify the level of P-selectin,IL-6 and TGF-β.Transcription levels of P-selectin and selectin dependent intercellular adhesion molecule-1(ICAM-1),IL-6 and transforming growth factor beta(TGF-beta)were tested with RT-PCR.Results:(1)The body weight of rats with diabetic nephropathy(DN)remained stable for groups of Fuc-50 and Fuc-200 compared with the group of Mod.Fucoidan could reduce urine volume in diabetic rats,especially in the high dose of Fuc-200.To a certain extent,fucoidan could inhibit the development and deterioration of DN.(2)Comparing with Nor group,urine volumes and M-TP in 24 h urine of DN rats was increased significantly.However,it could be reduced to the M-TP,UREA,u Alb,of urine and biochemical indicators of FRUC in blood,including the incidence of proteinuria in the fucoidan treated DN rats.That suggests that fucoidan can keep and protect the basic structure and function of glomerular,reducing renal damage caused by diabetes,and inhibiting the development of diabetic nephropathy.(3)It showed by the use of Hematoxylin-eosin staining(HE)that fucoidan could keep the normal physiological structure of the kidney in diabetic rats,reducing inflammation,and infiltration area of inflammatory cells.P-selectin and selectin depended inflammatory factors were down regulation in m RNA and protein levels significantly in kidney of DN rats for groups after the treatment of Fuc-50 and Fuc-200,suggesting that fucoidan could alleviate the inflammatory reaction of P-selection and inflammatory factor,which played a protective role on kidney function of diabetic rats.Conclusion:Fucoidan can alleviate polyuria,weight loss,renal damage and other typical symptoms of diabetes and diabetic nephropathy in DN rats,reducing the biochemical indicators of the urine and blood so as to protect and repair the normal physiological structure of glomerular inflammation,alleviating inflammatory reaction of P-selectin and Inflammatory factor,reducing the area of inflammatory cell infiltration,then the illness progression of diabetic nephropathy being impeded.Fucoidan can alleviate the inflammatory reaction of P-selection and inflammatory factor,which play a protective role on kidney function of diabetic rats.It has the potential to become a drug for the treatment of DN. |
