Diarrhea-predominant Irritable Bowel Syndrome(D-IBS),as a common clinical functional gastrointestinal disease,affects the quality of life of patients in many ways.However,the pathogenesis of the disease has not yet been fully elucidated.There are studies that indicate that irritable bowel syndrome has a close relationship with the variety of the changes of contentneurotransmitters in plasma,such as 5-hydroxytryptamine,vasoactive intestinal peptide,and substance-P.Western medicine treatment,such as antispasmodic agents,antidiarrheal,antidepressants,visceral analgesics,gastrointestinal microecological preparations and many more,has a certain degree of treatment on diarrhea-type irritable bowel syndrome.But as Chinese traditional medicine lovers,We hope to find and identify the cause and pathogenesis of the disease to explore the effective treatment of diarrhea-type irritable bowel syndrome,with the comparison of traditional Chinese medicine theory,in order to control the pathogenesis of the fundamental basis,and achieve the desired therapeutic goals.The thinking of this Chinese medicine prescription,based on the pathogenesis of MuyuShengfeng and ShiyunHuare,is called XifengHuashi.This Chinese medicine prescription is composed of Uncaria,Tribulus terrestris,Herbaceum AGLAOPHOTIS radix,Woody,Berberine,Gingiberi,Caluam,Patrina,Citrus,Ventus Inter Aquilonem,Atractylodes,Pelam and Salutem,et Licoricia.Although the clinical use of the side of the treatment of diarrhea-type irritable bowel syndrome was significant,the specific mechanism for its specific mechanism is not clear,and an experimental study is needed for further verification.Purpose:To investigate the role of changes in plasma and intestinal tissue neurotransmitter levels of 5-hydroxytryptamine,vasoactive intestinal peptide in the clinical symptoms of diarrhea-induced irritable bowel syndrome in rats.And to explore the treatment mechanism of the Chinese medicine prescription called XifengHuashi.Methods:Adult male Sprague-Dawley(Wistar)rats were divided randomly into normal control group,model group,high dose of Chinese medicine prescription group,medium dose of Chinese medicine prescription group,low dose of Chinese medicine prescription group and Piperidine group.In the model group,intraperitoneal acetic acid was stimulated once a day for 2 weeks from birth 8 days to 21 days.Normal group of rats daily rectal administration of saline 0.2?0.5ml.After the injection is complete,rinse the colon with 1 ml of 0.01 mol/L phosphate buffer(PBS).And than put the suckling rat into the cage free activities to eat water,suckling rats before leaving the mother live with milk.From 21d after 2wk,do not carry out any operation,but the normal feeding.The number of fecal points of defecation was observed at 14,15 and 16 days after the end of modeling.Observating the normal group at the same time.Abdominal retraction reflex(AWR)was evaluated after the end of the defecation statistics.Drug intervention was initiated on the 85th day of each group.Blank group and model group were given equal volume of distilled water gavage;In the low,medium and high dose groups of Chinese medicine,17,34,68g/(kg · d)were given according to the body surface area of 60 kg,the low,middle and high dose groups were the standard dose group and the dose group)Chinese medicine decoction;Western medicine group given Western medicine suspension.Continuous administration of 2 weeks,the average daily observation of animals,the day after the last administration of fasting blood and intestinal tissue,the determination of the indicators.The quantity of 5-hydrox-ytryptamine,vasoactive intestinal peptide,and substance-P in serum was detected by ELISA.And the quantity of 5-hydroxytryptamine,vasoactive intestinal peptide,and substance-P in intestinal tissue was determined by immunohitochemistry.Results:Blank group of mental state is good,shiny hair,fullness of the body.Chinese medicine high dose group of rats in general mental state,normal activity,hair color gloss no obvious abnormalities,moderate fullness of the body.Western medicine group and traditional Chinese medicine in the medium dose group of rats slightly awkward state of mind,burnout curl,coat color decreased,slightly thin body.Chinese medicine low dose group and model group rats mental state malaise,burnout curl,hair clutter,color dull,body weight loss obvious.The increase of traditional Chinese medicine concentration,the improvement was more obvious than model group(P<0.01)?and the effect of Chinese herbal decoction on the fecal water content was significantly improved.The effect of the middle dose group and the group was similar.The effect of high dose group was better than that of pinaverium bromide group(P<0.01).The content of serum neurotransmitter SP in Chinese rats was significantly improved compared with the model group(P<0.01).The effect of different concentration of Chinese herbal medicine on the changes of serum SP content in rats had a certain degree of correlation,but it was not statistically significant.Conclusion:The Chinese medicine prescription called XifengHuashi can improve the serum and intestinal tissue related neurotransmitter content,improve diarrhea-type irritable bowel syndrome in rats gastrointestinal function and visceral sensitivity,thereby improving the clinical symptoms of rats.
