Objective:Establish SD rat myocardial ischemia reperfusion(I/R)models.To research the effect of Dazhuhongjingtian injection on myocardial infarct size、the concentration of myocardial adenosine triphosphate(ATP)and the level of mitochondrial membrane potential(MMP)with ischemia-reperfusion rats.To explore the mechanism of Dazhuhongjingtian injection for the treatment of ischemic heart disease(IHD).Methods:40 male clean grade SD rats were randomly divided into sham operation group(group S),ischemia-reperfusion group(ggroup I/R),prevention group(group P),treatment group(group T)and prevention and treatment group(group P+T),each group averagely has eight rats.The rats of each group was respectively intraperitoneally injected with Dazhuhongjingtian injection by correspond requirements which is mentioned above.At the same time,establish the model of ischemia-reperfusion through the ligation of the left anterior descending(LAD)artery and removing the tubing.After the experiment was done,the heart was taken from the chest,and some fresh myocardial tissue was taken to determine the levels of ATP and MMP,and then the myocardial infarction area was calculated using Image J software.Results:1.The results of each group’s myocardial infarct size:Compared with group S,the myocardial infarct size of group I/S was larger with marked difference.(P<0.01);Compared with group I/S,myocardial infarct size of group P、group T and group P+T all got smaller with marked difference.(P<0.01);Among group P、group T and group P+T,there were no statistical difference(P>0.05);2.The concentration of each group’s ATP:Compared with group S,the ATP concentration of group I/S decreased significantly.(P<0.01);Compared with group I/S,the ATP concentration of group P、group T and group P+T all increased significantly.(P<0.01);The ATP concentration’s variation among group P、group T and group P+T was tiny and there’s no statistical difference among them(P>0.05);3.The level of each group’s MMP:Compared with group S,MMP level of group I/S dropped with marked difference.(P<0.01);Compared with group I/S,the MMP levels of group P、group T and group P+T all rised with marked difference.(P<0.01);The MMP level’s variation among group P、group T and group P+T was subtle and there’s no statistical difference among them(P>0.05);Conclusion:1.Dazhuhongjingtian injection could decrease the size of myocardial infarct with I/R rats and improve the prognosis.2.Dazhuhongjingtian injection could increase the concentration of myocardial ATP and the level of MMP,thus improve myocardial energy metabolism and promote the tolerance of myocardial ischemia.
