| Objective:To observe the protective effect of Rehmannia glutinosa decoction on H2O2-induced apoptosis of PC-12 cells,and explore the protective effect of Rehmannia glutinosa decoction on oxidative stress-mediated neuronal apoptosisMethods:PC-12 cells were treated with H2O2 to made to oxidative stress injury model.Use MTT to observe the effect of R.glutinosa decoction on the activity of PC-12 cells.The expression of Bax,Bcl-2 and cytochrome C,caspase3 expression in PC-12 cells were detected by Western blot.Results:The survival rate of PC-12 cells treated with 150μmol/L H2O2 was significantly decreased.After pretreatment with R.glurtinosa Decoction(2.5,5,10mg/mL),the cell survival rate was increased,approximately in a concentration-dependent manner(P<0.05).Significantly inhibited the expression of apoptosis-related proteins Bax,cytochrome C,cleaved-Caspase3,and increase the Bcl-2/Bax ratio(P<0.05).Conclusion:R.glutinosa decoction on the cells basically has no toxicity,and it can increase the expression of protective factor Bcl-2 in apoptosis and decreased the expression of apoptosis-promoting factor Bax.So the rate of Bcl-2/Bax is increased.It also can block the formation of mitochondrial membrane permeation channels,reduce the release of cytochrome C,ultimately reduce the activation of Caspase-3.It can protect neuronal mitochondria by regulating the expression of multiple apoptosis factors.In summary,R.glutinosa Decoction has protective effect on PC 12 cell apoptosis induced by oxidative stress.The protective mechanism is achieved by inhibiting the expression of apoptosis-related proteins Bax,cytochrome C,cleaved-Caspase3 and enhancing the expression of Bcl-2.This provides a strong laboratory basis for the application of R.glutinosa in clinical treatment on neurological diseases. |
