Microsphere as a slow controlled release drug carrier has become a hot research topic at home and abroad.In this paper,double emulsion evaporation method to prepare PLGA-Trail microspheres.The microspheres for drug loading,encapsulation efficiency,drug release,physicochemical properties and morphology,in vitro,investigated animal model experiment,the optimal preparation method in order to obtain further research,meet the demand.In the first part of this paper PLGA microsphere preparation method and research progress at home and abroad are reviewed,the microsphere preparation methods to conduct a comprehensive understanding and analysis,on this basis,this study determined using double emulsion evaporation technology for preparation of PLGA-Trail microspheres.The second part of the PLGA-Trail microspheres were fully optimized,and the physicochemical properties were investigated,the encapsulation efficiency and drug loading of microspheres as the evaluation index,and ultimately determine the PLGA-Trail microsphere preparation process is as follows: the concentration of PLGA is 120 mg/ml,the proportion of amount of water phase and oil phase1:20,water phase and oil ratio for the 1:20 and PVA was 5%,the vortex time is 90 s,the vortex velocity is 3000rpm/min,shear time 30 s,shear rate is 10000rpm/min.Again under the condition of PLGA-Trail microspheres prepared with good repeatability,encapsulation efficiency and drug loading of microspheres meet the experiment demand,encapsulation rate reached 51.7%,and particle size distribution in the 5.0-12.0μm range,arithmetic mean diameter was 9.3 + 4μm.The third part of the stability test of the PLGA-Trail microspheres,drug release in vitro and in vivo efficacy determination,The results showed that PLGA-Trail microspheres in vitro release drug release cycle is about 25d;that in vivo tumor bearing mice,PLGA-Trail microspheres can inhibit tumor growth,a single injection of PLGA-Trail microspheres in 21 d after tumor growth can be inhibited.In summary,the PLGA-Trail microsphere preparation process is reasonable,the microspheres with stable entrapment efficiency,drug loading content concentration inhibits tumor growth,the role of drug release significantly,with further research and development value.
