level.Functional food has good lipid-lowering and weight reduction research and development is a hot topic of concern.Recent research results indicate that the intestinal flora and the host.Diet interactions involved in the regulation of host energy absorption and metabolism,is the prevention and treatment of hyperlipidemia obesity "new target." in the Chinese herbal medicine tangerine peel is widely used and has a long history.Chen skin contains hesperidin,volatile oil,flavonoids and other active substances,can alleviate chronic inflammation,reduce blood glucose and body weight control effect.The previous work in our preliminary study confirmed that the use of continuous phase transition extraction technique from orange peel oil,can effectively regulate glucose metabolism by high fat diet induced by Or lipid metabolism,but it is not clear whether the imbalance of intestinal flora in tangerine peel can regulate the high fat diet caused.Therefore,this study intends to use metagenomic technology to explore the tangerine peel extract on high fat diet induced obese mice intestinal flora balance the effects of obesity on future research,provide a theoretical basis for the relationship with tangerine peel extract the intestinal flora.1.Peel extract to alleviate the symptoms of obesityThis paper studies the effect to relieve the symptoms of obesity in tangerine peel extract.4 week old female Kunming mice were randomly divided into normal group,high fat group,simvastatin group(2 mg/kg/d),tangerine peel extract high dose group(10g/kg/d),tangerine peel extract low dose group(5g/kg/d),8 rats in each group.The body weight of mice was observed every day the changes and related indices of obesity,and monitoring serum four blood lipids,blood glucose levels,lipoprotein lipase and hepatic lipase activity,to explore the role of lipid extracts of orange peel.The experimental results show that:compared with the normal group,high fat group body weight of mice was significantly increased in serum cholesterol and low density lipoprotein cholesterol were significantly increased(P<0.05),and serum lipoprotein lipase and hepatic lipase levels decreased.And different doses of simvastatin intervention or herbal extracts,can to some extent inhibit obesity.With the high fat group,tangerine peel mice weight,serum cholesterol,triglyceride,low density lipoprotein content decreased significantly(P<0.05),promote mouse serum lipoprotein lipase and hepatic lipase activity increased,in a dose-dependent manner.The lipid-lowering effect is better than that of group and simvastatin group tangerine peel extract.2.Based on high throughput technology,the effect of Pericarpium Citri extract on intestinal microflora in high fat mice was analyzedThis experiment was based on the Illumina MiSeq sequencing platform to carry out high flux sequencing of the DNA region of V3-V4 in high fat mice,and to analyze the effect of the extract of citrus peel on the diversity of intestinal flora in mice fed with high fat diet.Through OTU cluster analysis,five groups of mice 15 fecal samples were obtained 427080 qualified sequences.Dilution curve and rank OTUs number is 364,the normal group and the high fat group total OTUs number reduced to 196.The amount of OTUs increased in the high dose group and normal group,which showed that the intervention could improve the similarity of intestinal flora.242Analysis at the gate level:normal group mice feces,thick wall Firmicutes and Bacteroidetes were Bacteroidetes accounted for 46.66%of the total and 50.09%intestinal bacteria,Proteobacteria accounted for Proteobacteria compared with the normal group of 2.56%.,is still the dominant bacteria Firmicutes and Bacteroidetes in high fat mice in the gut,but the number of Firmicutes increased(62.28%vs 50.09%,P<0.05),the number of Bacteroidetes is relatively reduced(50.09%vs 34.95%).Simvastatin treatment group mice intestinal flora disturbance,but also has a number of Firmicutes,Bacteroidetes number(about 20%)trend.Contrast with the high fat group,tangerine peel extract intervention can significantly reduce intestinal Firmicutes number(P<0.05)compared with the normal group,the difference was not significant(P>0.05),and to a certain extent,promote the increased number of Bacteroidetes(34.95%vs 25.90%).In the analysis of department level:compared with normal group,high fat group of intestinal Lachnospiraceae,Lachnospiraceae(38%vs 25.539%)and Bacteroidales_S24 and 7_group(35%vs 7.99%)proportion decreased significantly(P<0.05),and the Department of rumen Bacteria(29.71%vs 6.88%)and Rikenellaceae(19.87%vs 2.73%)the proportion increased significantly(P<0.05).Compared with the high fat group,significantly increased the proportion of Lachnospiraceae mice by simvastatin group after treatment(41.65%vs 25.53%,P<0.05),almost restored to the level of control group.While the RIKEN strain ratio decreased significantly(19.87%vs 8.13%,P<0.05).After the intervention of tangerine peel extract,lachnospiraceae,almost recovered to the normal level.In summary,high fat diet damaged mice intestinal micro ecological environment,drug treatment group of simvastatin is effectively reduced the blood lipid indexes of blood glucose,but can not improve the disorder of intestinal flora.The tangerine peel extract intervention can not only relieve symptoms of obesity,while regulating the intestinal flora imbalance,specifically manifested in the diversity index of intestinal flora in mice.The composition
