Experimental Effect Of "Jiedu Huayu Jianpi Fang" On Expression Of P16、Thbs1、PTEN And E-cad Protein In Chronic Atrophic Gastritis With Dysplasia In Rats

Objective:To observe the effects of Jiedu Huayu Jianpi Fang on the expression of P16、thrombospondinl(Thbs1)、Phosphatase and Tensin homolog of chromosome 10 deletion(PTEN or MMACl、TEP1)and E-eadherin(E-cad)in rats with chronic atrophic gastritis complicating dysplasia.Methods:We use MNNG(N-methyl-N’-nitro-N-nitrosoguanidine)integrated modeling method to established the experimental rat model with chronic atrophic gastritis complicating dysplasia;the animal model rats were randomly divided into model control group,western medicine retinoic acid group and traditional Chinese medicine Jiedu Huayu Jianpi Fang group,and every groups were given the corresponding drug intervention,respectively;and the normal rats were selected as the positive control group;we application hematoxylin-eosin stainingtechnology to observe the morphological changes of gastric mucosa tissue in rats,and detected the expression level of P16、Thbs1、PTEN and E-cad in mRNA and protein level by immunohistochemistry、Real-time fluorescence quantitative polymerase chain reactionand Western blot method.Results:1.After eight weeks of experimental rats model build,build successful inspection after killing rats mode,drug intervention treatment,the pathology change.2.Through the Real-time PCR detection,the expression of p16、Thbs1、PTENand E-cad in mRNA decreased significantly in model group compared with blank group(P<0.01),Chinese traditional medicine group of P16,E-cad mRNA expression quantity is no statistical difference(P>0.05),Compared with model group,traditional Chinese medicine and western medicine group p16,Thbs1,E-cad mRNA expression of PTEN and E-cad increased significantly(P<0.01).Compared with western medicine group,Chinese medicine group of p16,Thbs1,PTEN and E-cad mRNA expression measuring tool was statistically significant(P<0.01).By Western blotting and immunohistochemical detection technology,compared with normal blank group,model group p16,Thbs1,PTEN and E-cad protein expression quantity also significantly lower(P<0.01);Chinese traditional medicine group of p16,Thbs1,PTEN and E-cad protein expression of statistically significant(P<0.05,P<0.01),in which the Western blotting detection p16,E-cad protein expression in the difference has no statistical significance(P>0.05).Compared with model group,traditional Chinese medicine and western medicine group p16,Thbs1,PTEN and E-cad obviously up-regulated cad protein expression(P<0.01).Compared with western medicine group,Chinese medicine groupof p16,Thbs1,PTEN,E-cad protein expression measuring tool was statistically significant(P<0.05,P<0.01).Conclusion:The therapy of Jiedu Huayu Jianpi could induce the protein expression increased of p16、Thbs1、PTEN and E-cad in chronic atrophic gastritis complicating dysplasia in rats,which could achieve the treatment of chronic atrophic gastritis complicating dysplasia and reverse the precancerous lesions of gastric cancer.
