OBJECTIVE:Intervene rats with chronic atrophic gastritis and dysplasia by Jianpi Qinghua Huayu Decoction,observe the changes of Jianpi Qinghua Huayu Decoction on the general condition of diseased rats and gastric mucosal damage,and detect NO,SOD,iNOS,IL in the gastric mucosa of rats The content of-1β and TNF-α and the changes in gene expression levels,to explore their targets for the treatment of chronic atrophic gastritis with dysplasia in gastric mucosal injury.METHODS:90 SPF male SD rats were randomly divided into 6 groups according to the random number table:normal group,model group,iNOS inhibitor group,low-dose Jianpi Qinghua Huayu Decoction group,middle-dose Jianpi Qinghua Huayu Decoction,high-dose Jianpi Qinghua Huayu Decoction group,15 rats in each group,Raised in 5 cages per cage.Except for the normal group,the other groups were given the SSI Sydney strain Hp suspension by gavage for 8 weeks.After the C13 breath test was performed,the infection was confirmed to be successful,and then the comprehensive modeling method(MNNG combined with ranitidine)was used for continuous modeling for 28 weeks.Randomly take 2 rats to observe the gastric mucosa every 8 weeks.After confirming the success of the model under light microscope,in addition to the normal group and the model group,the iNOS inhibitor group is given aminoguanidine by gavage,and the high,medium and low dose group of Jianpi Qinghua Huayu Decoction is given Different gradient concentrations of Jianpi Qinghua Huayu Decoction were taken after 10 weeks of intragastric administration.The gastric mucosa was observed under the HE staining light microscope.At the same time,NO and SOD kits were used to determine the NO content and SOD activity in the gastric tissue,respectively,and the real-time fluorescent quantitative PCR method was used to detect the changes in the expression levels of iNOS,IL-1β,and TNF-α genes.RESULTS:1.The effect of Jianpi Qinghua Huayu Decoction on NO and SOD in the gastric mucosa of rats:Compared with the model group,the NO content in gastric mucosa of the iNOS inhibitor group and the high and medium doses of Jianpi Qinghua Huayu Decoction group were significantly reduced,and SOD activity was significantly enhanced(P<0.05).The middle dose group of Jianpi Qinghua Huayu Decoction has the most obvious effect(P<0.01).2.The effect of Jianpi Qinghua Huayu Decoction on NO and SOD in the gastric mucosa of rats:Compared with the model group,the NO content in gastric mucosa of the iNOS inhibitor group and the high and medium doses of Jianpi Qinghua Huayu Decoction group were significantly reduced,and SOD activity was significantly enhanced(P<0.05).The middle dose group of Jianpi Qinghua Huayu Decoction has the most obvious effect(P<0.01).3.The effect of Jianpi Qinghua Huayu Decoction on the expression of iNOS,TNF-α,IL-1β in gastric mucosa of rats:Compared with the model group,iNOS inhibitor group and Jianpi Qinghua Huayu Decoction high and medium dose group gastric mucosa iNOS,TNF-αand IL-1 β was up-regulated(P<0.05),and the middle dose group of Jianpi Qinghua Huayu Decoction was the most obvious(P<0.01).CONCLUSION:Jianpi Qinghua Huayu Decoction can directly or indirectly regulate the expression of iNOS,reduce abnormal NO production,thereby reducing the oxidation,pro-inflammatory and cancer-promoting effects of NO on the gastric mucosa of rats,and ultimately improve the inflammation,atrophy,and dysplasia of the gastric mucosa in rats,Prevent cancer.
