Objective To explore the effect and mechanism of GOLPH3 gene silencing on invasion and migration of gastric cancer cells.Methods The lentivirus containing GOLPH3 siRNA sequence andnegative control sequence were infected into gastric cancer cell lines SGC-7901,respectively. Fluorescence microscopy was used to detect the expression of green fluorescent protein(GFP). GOLPH3 down regulation was verified by quantitative real-time PCR and Western blotting. Cell invasion and migration were affirmed by matrigel invasion assay and transwell migration assay. The expression of mTOR,p-mTOR ,STAT3,p-STAT3 protein was affirmed by Western blotting.Result Gastric cancer cells expressing high levels of GFP were successfully obtained, with an infection rate of approximate 90%. GOLPH3 expression was significantly down-regulated in SGC-7901 cells infected with the lentivirus containing the GOLPH3 siRNA compared with SGC-7901 cells transduced with the empty vector or noninfected SGC-7901 cells(P<0.05 for both) . Invasion assay displayed that the numbers of cells passed through matrigel membrance were significantly lower in GOLPH3-siRNA group(70.3±8.5) than negative control group(171.67±3.2) and blank control group(167.6±11.0).Migration assay displayed that the numbers of cells passed through polycarbonate filters were significantly lower in GOLPH3-siRNA group (82.0±3.6) than negative control group(177.0±5.29) and blank control group(174.3±4.7).The expression of p-mTOR、 p-STAT3 protein in GOLPH3-siRNA group were lower than the negative control group and blank control group with statistical difference(all P<0.05),but the expression of mTOR、STAT3 protein were not changed obviously in all the groups.Conclusion 1. Silencing GOLPH3 gene could inhibit the invasion and migration of gastric cancer cells; 2. Silencing GOLPH3 gene could down regulate the protein expression of p-mTOR、p-STAT3.
