Objective:In this study,we used Real Time Reverse Transcriptase PCR(qRT-PCR)to detect the expression of DOG1 in peripheral blood mononuclear cell(PBMC)of Gastrointestinal stromal tumor(GIST)patients,and to analyze the relationship between the expression of DOG1 in PBMC and the clinicopathological factors and prognosis,At the same time,we also Preliminary discuss the feasibility of DOG1 as a serum molecular marker of GIST.Materials and Methods:Randomly selected from December 2015 to December 2016 in the Second Affiliated Hospital of Nanchang University and in line with the Chinese gastrointestinal stromal tumor diagnostic criteria for the diagnosis of GIST of 26 patients as the experimental group,while randomly selected the same period of 36 healthy volunteers with the same characteristics as the experimental group as the Control group,and then collected its peripheral venous blood samples and clinical pathology datas.The peripheral blood mononuclear cell were isolated by density gradient centrifugation,and the total RNA was extracted and then reverse transcribed to synthesize c DNA.The expression level of DOG1 was detected by qRT-PCR,by which the Ct value of DOG1 was calculated,and the relative expression of DOG1 was calculated by relative quantitative 2-ΔCt method.The expression level of DOG1 in peripheral blood of both group were observed and compared,and the relationship between the expression of DOG1 in PBMC and the clinicopathological factors and prognosis were analyzed.The receiver-operating characteristic curve(ROC curve)was made according to the expression level of DOG1 in peripheral blood,the sensitivity and specificity of DOG1 as GIST serum markers were analyzed to find the best critical point.All of the datas were analyzed by SPSS19.0 software and plotted with GraphPad Prism 5.0 software.Results:1、The mean expression level of DOG1 in peripheral blood of 26 cases of GIST was 3.800 ± 2.213,and the mean expression level of DOG1 in 36 healthy controls was 0.909 ± 0.921.The difference of the mean expression level of DOG1 in peripheral blood between the two group was statistically significant(P <0.01).2、The expression of DOG1 in PBMC of GIST was not correlated with gender,age,tumor size(diameter),primary tumor site,number of tumor mitochondria and risk grading(P >0.05),but related to the expression of Ki-67 Index of tumor cells(P <0.05).3、According to the ROC curve,when the expression level of DOG1 was at 1.307,the sensitivity and specificity of diagnosis were the highest,also,the diagnostic value was the highest(the sensitivity and the specificity were 88.5% and 88.9%,respectively),and the AUC was 0.890(95% CI,0.798 ~ 0.983,P <0.001).Conclusion:1、 The expression level of DOG1 in peripheral blood mononuclear cell of GIST is significantly higher than healthy people.2、The expression of DOG1 in PBMC of GIST was not correlated with gender,age,tumor size(diameter),primary tumor site,number of tumor mitochondria and risk grading(P >0.05),but related to the expression of Ki-67 Index of tumor cells(P <0.05).3、The expression of DOG1 in PBMC of GIST may be related to prognosis.4、DOG1 is expected to be a serum molecular marker for the diagnosis of GIST.
