Schizophyllum commune belongs to the genus Schizophyllum of the family Schizophyllaceae in the Agaricales orde.It is also called tree-flower,white-flower,etc.And it is a kind of edible and medicinal mushroom which contains many biological functions including keeping body strong,anti-tumor,treating neurasthenia and dizziness tinnitus.Some compounds with anti-tumor,anti-inflammatory and antimicrobial activities have been identified from extracts of S.cowirmmune.But the contributors and how to contribute for anti-pain is unknown in S.commune.So it should be worthy of further research to provide guidance to find new lead compounds.In this study,a variety of chromatographic techniques(the thin layer chromatography(TLC),tosilica gel column chromatography,sephadex LH 20,and reverse-phase high performance liquid chromatography(RP-HPLC),etc.)and identification technologies(1H-NMR,13C-NMR,ESI-MS,etc.)were used to isolate,purify and identify the S.commune.And we used patch clamp to track the activity on NaV channels of isolation of fractions.We also tested the compounds from S.commune in two kinds of mice pain models in which pain was induced by noxious chemicals.The main results as follows:1.Compounds from S.commune were isolated and identified:Five phenolic acids were identified from extracts of the fungus of S.commune.They are vanillic acid(Ⅰ),m-hydroxybenzoic acid(Ⅱ),o-hydroxybenzeneacetic acid(Ⅲ),3-hydroxy-5-methybenzoic acid(Ⅳ)and p-hydroxybenzoic acid(Ⅴ).All of these compounds were first reported from S.commune.2.The five compounds effect on NaV channels:They show inhibitor activity against both tetrodotoxin-resistant(TTX-R)and tetrodotoxin-sensitive(TTX-S)NaV channels.All compounds show low selectivity on NaV channel subtypes.3.Three phenolic acids effect on mice pain models:After intraperitoneal injection,three compounds(p-hydroxybenzoic acid,vanillic acid and 3-hydroxy-5-methybenzoic acid)exert analgesic activity in mice models significantly.Through the experiment we confirmed that the S.commune has analgesic active compounds.As a medicinal fungus,S.commune has been used for treatment of neurasthenia which caused by muscle pain,pain,dizziness,tension headaches and so on.We think these compounds which first isolated from S.commune may play an important role to treat neurasthenia.This study not only provides a possible mechanism how S.commune treats of neurasthenia,but also has far-reaching significance to complete active component of S.commune.What’s more,it provides guidance to find new lead compounds for clinical pain management.
