ObjectiveTo observe the antitumor synergic effect of Qijiaoshengbai Capsule in combination with Cyclophosphamide on S180 tumor bearing mice and its related mechanism.MethodsMouse S180 cells were subcutaneously injected into right axillary region of the mice to establish S180 tumor-bearing models.One hundred and five mice were randomly assigned into control group,Low dose group of Qijiaoshengbai Capsule(0.5g/kg),Middle dose group of Qijiaoshengbai Capsule(1.0g/kg),High dose group of Qijiaoshengbai Capsule(2.0g/kg),CTX(30mg/kg)group,Low dose Qijiaoshengbai Capsule(0.5g/kg)combined with CTX group,Middle dose Qijiaoshengbai Capsule(1.0g/kg)combined with CTX group,High dose Qijiaoshengbai Capsule(2.0g/kg)combined with CTX group,respectively had drug administration for 15 consecutive days,daily measuring the weight and tumor volume of each groups,24 hours after last administration take the blood of tumor-bearing mice for blood test,dissect the body and take the tumor,spleen and thymus out to calculate tumor inhibition rate,thymus index and spleen index,take the liver and kidney out for H&E staining to observe the injury state.In the aspect of mechanism,we extracted tumor cells from tumor tissues and applied Annexin ? /PI flow cytometer method to detect the apoptosis of tumor cells condition,Western-blot method to detect the expression of p53 and the bcl-2 in tumor tissue,and applied immunohistochemical method to detect the expression of VEGF and CD34.ResultsQijiaoshengbai Capsule in different dosage groups can increase the tumor weight and tumor inhibition rate of tumor bearing mice,especially in QSC middle dose group,and also the apoptosis rate of tumor cells in each dose group is higher than in the model group,we can include that QSC had a certain inhibitory effect on the growth of tumor in S180 tumor bearing mice.Compared with group CTX,after the end of administration,the body weight of mice in each doses of QSC combination groups is increased and the relative tumor volume of mice is lower than CTX group,also as the tumor weight.The anti-tumor rate and apoptosis rate of the combined dose groups were higher than those in the CTX group,which indicated that the combination of QSC and CTX could obviously inhibit the growth of transplanted S180 tumor in mice.We used QSC combined with CTX and research the toxicity of chemotherapy induced by CTX,and found that the degree of hematotoxicity were decreased,the number of blood cells increased,liver and kidney damage were reduced.The apoptosis rate of tumor cells was significantly increased and synergistic effect was obviously.In the research of mechanism,Qijiaoshengbai Capsule in combination with CTX could be reduced p53,bcl-2 protein levels,and reduced the expression of CD34,VEGF in tumor tissues,the effect of Middle dose combination group was most significantly.ConclusionThe Qijiaoshengbai Capsule had a certain inhibitory effect of S180 tumor growth in vivo,and in combination with Cyclophosphamide made the antitumor effect more obvious in synergy and attenuation.And the antitumor effect was related to the expression of tumor apoptosis regulatory proteins p53,bcl-2 and the microvessel density control proteins CD34,VEGF.
