Objective:Study on the protective effect and mechanism research of astaxanthin on reproductive damage male rat induced by ornidazole,and to provide a theoretical basis for clinical treatment of infertility.Methods:40 adult male SD rats were randomly divided into 5 groups of 8 animals each,and received medication orally for 3 weeks.Group A rats received solvent(0.5%carboxymethylcellulose solution + 1 ml com oil),group B rats received ORN 400 mg/(kg·d),group C rats received ORN 800 mg/(kg · d),group D rats received ORN 400 mg/(kg · d)?AST 20 mg/(kg · d),group E rats received ORN 800 mg/(kg · d)+ AST 20 mg/(kg · d).Recorded the general situation,24 h after the last administration,rats were anesthetized by intraperitoneal chloral hydrate.Rat testis,epididymis and seminal vesicle were removed and weighed,measured the sperm concentration and activity of one side epididymal tail,the remained epididymal and one side testis were histopathologically examinated;detected GSH-Px,GR,CAT,SOD activity and MDA content of other side epididymis tissue;detected the LDH,SDH,y-GT activity and 8-OHdG level of other side testis tissue.Results:1.During the experiment,the activity of group A rats were normal,the weight was increased obvious and the fur was shiny.Group B,C gradually appear listlessness,slow reaction,drowsiness and other symptoms,and was worse with the time of administration.2.The results of HE in testis and epididymis showed that the spermatogenic cells in seminiferous tubules of A group were arranged in neat and tidy,with clear structure,dense and uniform nuclear staining;a large number of linear sperm were seen in the lumen of the epididymal duct,cilia arranged neatly.The morphology of testis and epididymis of group B and D were almost no difference compare with,group A.The spermatogenic cells in seminiferous tubules of group C were significantly decreased,hierarchical was fuzzy,intercellular space becomes biger and the vacuole increases;the number of sperm in the lumen of the epididymal duct was significantly reduced,the arrangement was loose and disordered,there were a variety of nonsperm cell components and large blank areas.Testicular tissue structure of group E was restored a lot and epididymal duct sperm was increased,but still worse than group A.3.Compared with group A,epididymal sperm motility,epididymis GSH-Px,SOD activity of group B decreased significantly,the content of MDA increased significantly(P<0.05),Other changes were not obvious;epididymal sperm motility and concentration,index of testis,GSH-Px,GR,CAT and SOD activity of epididymis tissue homogenate,LDH,SDH,y-GT activity of testis tissue homogenate in group C were significantly decreased,while the content of MDA of epididymis and 8-OHdG level of testis increased significantly(P<0.05).After AST treatment:compared with B group,the epididymis sperm and SOD activity increased significantly of group D,while the content of MDA decreased significantly(P<0.05);compared with group C,the weight gained,the sperm motility and LDH,SDH activity of testis was increased significantly of group E,while the content of MDA decreased significantly(P<0.05);Other indicators improved but no statistical difference.Conclution:AST can improve the sperm quality of reproductive damage rats induced by ORN,especially sperm motility;The mechanism may be related to the increase of antioxidant capacity of the rat epididymis,the activity of testicular mark enzyme and the inhibition of NDA damage of testis cell.
