BackgroundFunctional dyspepsia(functional dyspepsia,FD)is a common and frequent clinical disorder of gastrointestinal function,main show is Postprandial full bilge unwell,early enough,upper abdominal advantages in the treatment of FD,this study by adopting the spleen prescription intervention by way of in vivo and in vitro FD spleen deficiency rat model of combined disease and study its combined diseaseObjective:The establishment and evaluation of FD spleen deficiency rats combined disease modelMaterials and methods:Will be 30 male SD rats(rats)were randomly divided into normal group,the pure iodine acetamide lavage group(disease),iodine acetamide lavage superposition small platform standing group(group)combined disease,born 7 purchases,feeding adaptability to 3 days.Born 10 ConditionBefore the treatment,compared with normal group,model group and each group of rats body weight,food intake,water content,holding power were significantly lower(P<0.05).Compared with model group,in addition to the normal group,the rest between each rat body weight,food intake,water content,holding power had no obvious difference(P>0.05).After treatment,compared with model group,western medicine group,the sand is low,medium and high dose group,pixu(spleen deficient),high dose group of rats body weight was significantly increased(P<0.05).Sand high dose group,high dose group of rats drinking pixu(spleen deficient)significantly increased(P<0.05).Sand high,medium and low dose group,high,medium and low dose group of pixu(spleen deficient)significantly increased food intake in rats(P<0.05).Western medicine group,and sand high,middle dose group,high and low dose group of pixu(spleen deficient)holding power of rats significantly increased(P<0.05).Compared with western medicine group,sand high dose group,high dose group of rats drinking pixu(spleen deficient)significantly increased(P<0.05).High,middle dose group of pixu(spleen deficient)rats significantly increased food intake(P<0.05).Each rat body weight,holding power difference compared with the western medicine group had no statistical significance(P>0.05).2 Stomach retention rate and small intestine propulsion(1)compared with normal group,model group rats gastric residual rate is significantly higher than in normal group(P<0.01);Compared with model group,western medicine group,low dose pixu(spleen deficient),sweet sand high dose group of rats gastric residual rate significantly decreased(P<0.05);Compared with western medicine group,sand of pixu(spleen deficient),high dose group,low,middle dose group of rats with gastric residual rate is significantly higher than in western medicine group(P<0.05).(2)compared with normal group,model group in the rat small intestine propulsion ratio was significantly lower than normal group(P<0.05);Compared with model group,western medicine group,the low,middle and high dose group of pixu(spleen deficient),sand high dose group of small intestine propulsion ratio significantly increased(P<0.05);Compared with western medicine group,high dose group of small intestine propulsion pixu(spleen deficient)were significantly higher than western medicine group(P<0.01),and sand low doses of small intestine propulsion ratio was significantly lower than western medicine group(P<0.05).3 Serological Indicator(1)compared with normal group,model group,the serum D-xylose content was significantly lower than normal group(P<0.05);Compared with model group,western medicine group,low dose group of serum D xylose content pixu(spleen deficient)were significantly higher than that of model group(P<0.01);Compared with western medicine group,the serum D-xylose content differences between traditional Chinese medicine group were not significant(P>0.05).(2)compared with normal group,model group was significantly reduce serum amylase content(P<0.05);Compared with model group,western medicine group,high dose group,pixu(spleen deficient)and the sand is low,serum amylase content in high dose group were significantly higher than that of model group(P<0.05);Compared with western medicine group,the serum amylase content differences between traditional Chinese medicine group were not significant(P>0.05).(3)compared with normal group,model group significantly increased serum lactic acid content in the model group(P<0.05);Compared with model group,western medicine group,after the treatment of pixu(spleen deficient),high dose,the sand is low,the dosage of serum lactic acid levels were significantly lower than model group(P<0.05);Compared with western medicine group,the serum lactic acid content differences between traditional Chinese medicine group were not significant(P>0.05).(4)compared with normal group,model group serum gastric dynamic content was significantly lower than normal group(P<0.01);Compared with model group,western medicine group,the low,middle and high dose group of pixu(spleen deficient),the sand is low,the serum gastric dynamic content,high dose group were higher than that of model group(P<0.01);Compared with western medicine group,sand of pixu(spleen deficient),high dose group,low serum gastric dynamic content,middle dose group were higher than that of western medicine group(P<0.05).(5)compared with normal group,model group serum gastric secrete element content is significantly lower than normal group(P<0.01);Compared with model group,western medicine group,the low,middle and high dose group of pixu(spleen deficient),incense in the sand,low serum gastric secrete element content,high dose group were higher than that of model group(P<0.01);Compared with western medicine group,high dose group of pixu(spleen deficient)serum gastric secrete element content concentration,and the difference was statistically significant(P<0.05).(3)provide amplitude with concentration and dosage of gastric antrum after enhanced obviously,the difference was statistically significant(P<0.05),the contraction frequency no significant change(P>0.05),give sweet sand 6 gentleman when low concentration can significantly increase the amplitude and frequency of gastric antrum(P<0.05),when the concentration reaches a certain dosage is inhibition of gastric antrum contraction frequency and amplitude(P>0.05).(4)after gastric body contraction amplitude decreases with drug concentration,the difference was statistically significant(P<0.05),the contraction frequency is increased,the difference was statistically significant(P<0.05),give sweet sand 6 gentleman amplitude and frequency of gastric body when different concentrations were significantly decreased,the difference was statistically significant(P<0.05),when the concentration reaches a certain dosage observing different spleen medicine on in vitro amplitude and tension of stomach bottom of gastric antrum of rats,to explore in vitro under different spleen medicine for stomach dynamic adjustment.Materials and methods:Experimental animals can be divided into four groups,taking gastric antrum of rats,stomach bottom different organizations,in vitro to different concentrations of spleen medicine,medicine of down-regulated,spleen wet medicine and traditional Chinese medicine drug combinations,observation on amplitude and the role of stomach bottom tension difference of gastric antrum.Results:(1)traditional Chinese medicine spleen,spleen wet group and can significantly improve the tension of stomach bottom in the group,the difference was statistically significant(P<0.05),down-regulated group can significantly reduce the tension of stomach bottom,the difference was statistically significant(P<0.05).(2)Chinese medicine spleen group,spleen wet group can increase gastric antrum contraction,the difference was statistically significant(P<0.05),down-regulated group can significantly reduce the distal gastric contraction,the difference was statistically significant(P<0.05),group of gastric antrum in spleen temperature effects have no difference(P>0.05).Conclusion1.Iodine acetamide lavage superposition method improved small platform standing to make animal model in line with the characteristics of spleen deficiency,functional dyspepsia with gastric dynamic disorders.2.The spleen prescription can improve FD rats gastrointestinal absorption function and lactic acid accumulation caused by limb tired fatigue state.3.The spleen prescription can directly promote the gastric emptying and small intestinal propulsion in rat models or by improving the gastrointestinal hormone,adjust gastric bowel movement,among which side of the small intestine propulsion function I pixu(spleen deficient)is superior to the sand 6 gentleman.4.In the condition of in vitro,spleen and prescription can be directly ACTS on the stomach tissue and improving stomach,spleen and spleen medicine prescriptions to exert its from different angles of gastrointestinal two-way adjustment function,and present a dose-dependent.
