| Micellar liquid chromatography(MLC)is a new mode of reversed-phase liquid chromatography with aqueous solution of surfactants above the critical micellar concentration(CMC).The system provides hydrophobic and electrostatic sites of interaction for solutes to allow an effective separation of compounds of different natures.Biopartitioning micellar chromatography(BMC)is a mode of MLC,which uses polyoxyethylene(23)lauryl ether(Brij 35)solution above CMC as mobile phases under physiological conditions(pH,osmotic pressure,temperature,etc.).BMC’s system could describe the biological behavior of drugs and simulate biopartitioning process.The topic is aimed to study the application of MLC,especially the application of BMC in the pharmaceutical analysis.In the first part,the capability of BMC to describe and estimate bioactivity data of anticancer drugs has been studied.The retention of 17 anticancer drugs was studied using different pH of Brij-35 as micellar mobile phase in modified C18 stationary phase.Quantitative retention-activity relationships(QRAR)of half-life time(t1/2),area under concentration-time curve(AUC),clearance(CL),volume of distribution(Vd)and the toxicity(LD50)were investigated for these compounds.The results indicated that using the one descriptor(log k)to describe the bioactivity of anticancer drugs is feasible.In the second part,the complex ingredients in traditional Chinese medicine(TCM),Xiaoyanlidan tablets for example,were separated by BMC,and the aim of this part is to estimate the permeability of each ingredients.The chromatographic conditions were investigated to achieve a good separation,and quantitative retention-permeability relationships(QRPR)models were established to estimate the permeability of components in Xiaoyanlidan tablets under this conditions.In third part,the simultaneous determination of andrographolide and dehydroandrographolide in Xiaoyanlidan tablets by MLC was studied.The separation characteristics of solutes were investigated in kinds of surfactants,concentration of micellar solutions,and ratio of organic additives.The method validation showed that the method was simple,reliable,and provided a reference standard for the quality control of TCM. |
