Gentiana macrophylla Pall,which belongs to Gentiana genus of Gentianaceae family,is a kind of traditional folk medicine which is famous for treating rheumatism.The chemical constituents of Gentiana macrophylla Palll.include iridoids,triterpenoids,flavonoids,alkaloids,and other types of secondary metabolites.Little is known about constituents and biological activities of Gentiana macrophylla Pall.The purpose of this research is mainly to separate n-butanol extract layer of the ethanol extract of Gentiana macrophylla Pall.The aim of this study was to get new compounds by different kinds of extraction technique,subjecting to repeatedly different kinds of column chromatography and semi-preparation high performance liquid chromatogra,And by the application of NMR,and HR-ESI-MS spectroscopy detection technology,about this research 20 compounds were identified,Including,gentiana ?(1),gentiana?(2),swertiamarin H(3),3-O-Caffeoylgentiopicroside(4),5-ethylidence-3,4,5,6-tetrahydro-cis-6,8-dimethoxy-1H,8H-pyrano[3,4-c]pyran-1-one(5),gentiopicroside(6),swertiamarin(7),sweroside(8),ethyl-3-(?-D-glucopyranosyloxy)-2-hydroxybenzoate(9),blumenol C(10),byzantionoside B(11),2-Phenylethyl-?-D-glucopyranoside(12),(+)-Syringaresinol-?-D-glucoside(13),(-)-syringaresinol-4,4'-bis-?-O-D-glucopyrannoside(14),4',5,7-trihydroxyflavone(15),5,7-Dihydroxy-4'-methoxyflavone(16),?-daucosterol(17),?-sitosterol(18),dibutylphthalate(19),?-D-fructofuranosyl-a-D-glucopyranoside(20),?-D-glucofuranosyl-a-D-glucofuranosl-?-D-glucofuranoside(21),gentianose(22).Compounds 1,2,3,4,9 are new and Compounds 10,11,12,21 are firstly isolated from the Gentianaceae family.Nitric oxide(NO)is an important mediator in the living organism that is synthesised from L-arginine using NADPH and molecular oxygen.However,excessive production of NO catalysed by inducible NO synthase is pathogenic for host tissue.Thus,effective inhibition of NO accumulation represents a beneficial therapeutic strategy for the treatment of NO-mediated disorders.In this article,we are describing the isolation and identification of 11 compounds from Gen.tiana macrophylla Pall,and their NO inhibitory activity in lipopolysaccharide(LPS)induced NO production in RAW264.7 cells.All of the constituents showed dose-dependent nitric oxide(NO)inhibitory activity against lipopolysaccharide-induced NO production in RAW 264.7 macrophage cells suggesting their possible use as anti-inflammatory agents.
