| The research project is part of the National Natural Science Foundation –“Study the Pharmacodynamic material basis for the Smilax macrophylla anti-gouty arthritis and its in vivo pharmacokinetics”(No.30960494),the subject is Smilax macrophylla.Smilax macrophylla is the root of Smilax microphylla C.H.Wright which is the part of Smilacacea.Smilax microphylla,familiarly known as “wu yu ci” and“di fu ling teng”,is a creeping,extremely tough shrub of the family Smilacaceae.It grows in bushy places and in wood land,forming frequently hedgerows and mainly distributed in Eastern and Southern China.Sweet,bitter,flat,documented the effectiveness of chills and active,expectorant cough,mainly used for clinical treatment of gouty arthritis,wound,lumbar muscle strain,bronchitis,tuberculosis,cough,hemoptysis and other diseases.Related literatures have been reported that as a Chinese herbal medicine,Smilax,generally have anti-inflammatory effects,and treatment of arthritis has a significant effect.The modern pharmacological studies have shown that taking the Smilax macrophylla extract orally can significantly promote the excretion of uric acid of the disease model rats which have high blood uric acid to be a lower level in the blood.However,to our best knowledge,there has been no detailed report on the pharmacodynamic material basis and mechanism of action in vivo of Smilax macrophylla.1.Studies on the Chemical Constituents in Ethyl Acetate Parts of Smilax macrophyllaThe ethyl acetate parts of Smilax microphylla were investigated on the chemical constituents systematically by various kinds of chromatographic methods.The structure of ten compounds was determined by means of chemical evidence and spectroscopic analysis.These compounds are 1 lignans,2 sterols,3 steroidal saponins,3 stilbenes.They are listed as follows: lyoniresinol-9-O-8’’-syringylglycerol ether(E3-5),Quercetin-3’-glucoside(E6-2),β-sitosterol(E2-1and PD-1),Daucosterol(E4-2),Diosgenin(E2-3)、Protodioscin(E6-11)、Prosapogenin B of dioscin(E5-3)、Piceatannol(E4-3)、Resveratro1(E5-2)、Oxyresveratrol(E5-1).Among the ten compounds,a new lignan,lyoniresinol-9-O-8’’-syringylglycerol ether was a new compound,and the others were isolated from Smilax microphylla for the first time.2.The study of the fingerprint on six plants in SmilacaceaThe chromatogram of six plants in Smilacacea were determined by HPLC,with the investigation of the extraction conditions,chromatographic conditions,methodology,and the method is reliable and good reproducibility.The similarities and differences of the six plants were contrasted by fingerprinting analysis method.It partially revealed the identification of different plants and supplied a reference of quality control in Smilacacea,and it also provided the scientific basis for the fingerprint of Smilax macrophylla.In short,through the studies mentioned above on Smilax macrophylla it can provide the reference for the identification of Smilacacea.Our study provided the theoretical basis for further study,development and application of Smilax macrophylla. |
