Objective:To investigate the effect of epidermal growth factor(EGF)on the proliferation of endometrial adenocarcinoma cells,phosphorylation of estrogen receptor α(ERα)and Ack1 in the absence of estrogen.Methods:1.Ishikawa cell line was stimulated by EGF and tyrosine inhibitor dasatinib without estrogen settings,Cell Counting Kit-8(CCK-8)was used to evaluate cell proliferation,statistical software SPSS19.0 was used for data analysis.2.Ishikawa cell line was stimulated by EGF and tyrosine inhibitor dasatinib without estrogen settings,Western blot was used to detect ER α phosphorylation and Ack1 phosphorylation,Image J image analysis software was used to analyze the relative content of the target protein.Results:1.EGF promotes the proliferation of Ishikawa cells in the absence of estrogen,compared with the control group,there was significant differences,P<0.05.2.Dasatinib inhibited the proliferation of Ishikawa cells induced by EGF,compared with the EGF treatment group,there was significant differences,P<0.05.3.EGF induced Ack1 and Tyr-537 ERα phosphorylation of Ishikawa cell lines.Compared with the control group,the phosphorylation level of Ack1 and Tyr-537 ERαwere increased,there was significant differences,P<0.05.4.Dasatinib inhibited EGF induced phosphorylation of Ack1 and ERα Tyr-537.When the concentration of Dasatinib was higher than 400 nmol/L,the phosphorylation of Ack1 and Tyr-537 ERα was inhibited completely,compared with the control group,there was significant differences,P<0.05.Conclution:1.EGF can promote the phosphorylation of Ack1 kinase and ERα Tyr-537 in endometrial adenocarcinoma Ishikawa cells and promote the proliferation of Ishikawa cells(P<0.05).EGF has a significant role in the development of endometrial cancer cells,which provides a theoretical basis for the search for novel anticancer drug therapeutic target.The aim of this study is to provide a theoretical basis for the treatment of new anticancer drugs.2.Dasatinib inhibited EGF induced phosphorylation of Ack1 and ERα Tyr-537,and inhibit the proliferation of Ishikawa cells,Dasatinib will be used in the treatment of endometrial carcinoma.
