The Study Of GRM5 Express On Glioma And Correlation With Glioma U251 Cell Ability Of Migration And Invasion | | Posted on:2018-01-22 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y S Li | Full Text:PDF | | GTID:2334330533465549 | Subject:Neurosurgery | | Abstract/Summary: | PDF Full Text Request | | Objection:1.Study metabotropic glutamate receptor5(GRM5)express on human glioma and correlation with Pathologic gradation of glioma.2.Study of GRM5 Impact on U251 cell ability of migration and invasion.Methods:1.In order to study the glioma tissues GRM5 expression,We collected in our hospital neurosurgery,13 cases of gliomas specimens,All specimens according to world health organization(World Health Origanization,WHO)central nervous system tumor classification criteria for pathological diagnosis and grading in 2007.Take the method of immunohistochemical detection GRM5 expression in glioma specimens.2.In order to study the GRM5 affect glioma U251 cells’ ability to migrate,We de signed and synthesized two specificity for GRM5 genes of smallinterferingRNAs(GRM5-si RNA-1247,GRM5-siRNA-1587)and control group RNA(GRM5-siRNA-NC).Transf ectionU251 cell lines after 48 h using Western blot(protein imprinting method)to det ect GRM5 expression.3.Made transfection exeriments and Tanswell experiment after U251 cells transfection.Validation siRNA(GRM5-siRNA-1247,GRM5-siRNA-1587),The influence of the migration and invasion ability of U251 cells after nterference GRM5 expression.Reasults1.Immunohistochemical results show GRM5 in low level and high level are expre ssed in the glioma.And along with the increased level of tumor,Express quantity also increased,The results have statistical significance(P<0.01).2.Western Blot experiments results show that the U251 cells GRM5 expression q uantity is decreased obviously after transfection siRNA(GRM5-siRNA-1247),The result s have statistical significance(t=3.020,P<0.05).3.The scratches experiment result show that Cell scratch wound healing rate signi ficantly lower than the control groupn after U251 cells transfection siRNA(GRM5-siR NA-1247),The results have statistical significance(24h t=6.751,P<0.01;48h t=8.668 P<0.01).4.Tanswell experimental results show that Cell invasion ability significantly lower than the control group after U251 cells transfection si RNA(GRM5-siRNA-1247),The results have statistical significance(t=20.726,P<0.01).Conclusion:1.GMR5 have expressed in the glioma,and increased with the level of glioma,i ncreased expression quantity.2.GRM5 can promote Glioma U251 cell the ability of migration and invasion. | | Keywords/Search Tags: | Glioma, GRM5, siGRM5, Migration, Invasion | PDF Full Text Request | Related items |
