| Objective: Investigation of effects of female ovarian function and gonadotropin on aneuploidy of oocytes.Establishment of stable human oocyte preparation and fluorescence in situ hybridization technology platform.Methods: Non-fertilized oocytes was obtained from patients who had undergone an IVF / ICSI cycle from October 2016 to December 2016 in the First Hospital of Lanzhou University.The oocytes were immobilized and examined by fluorescence in situ hybridization(FISH)to detect the aneuploidy of chromosome 13,16,21 and 22 after authorization.The incidence of aneuploidy were compared with female age,antral follicle count(AFC),anti-Mullerian hormone(AMH),basic follicle stimulating hormone(bFSH)and the starting dose of follicle stimulating drug,the usage days,the total dose of drug in assisted reproductive technology to explore the relationship.Results: A total of 257 in 347 non-fertilized oocytes obtained were successful immobilized on 62 pieces of microslide.220 non-fertilized oocytes were identified with a clear chromosomal structure and recognizable signals under a fluorescence microscope.The detection rate of aneuploidy in oocytes was 17.27%.Comparison of clinical data found that,at the overall level,the aneuploid oocyte group has higher age,AFC and serum AMH than other oocyte groups,the difference is statistically significant(P <0.05).The mean serum FSH of the aneuploid oocytes was greater than that of the other oocytes,but the difference was not statistically significant.After classified by age,there was no significant difference in AFC,serum AMH and serum FSH in the groups under 35 years of age.In the 35 and older group,AMH was lower in the aneuploid oocyte group,the difference was statistically significant,but the difference of AFC and FSH was not statistically significant.There was no statistically significant difference between protocols of ovulation stimulation.According to the protocol grouping,the starting dose of follicle stimulating drug,the usage days,and the total dose of drug had no significant differences.Conclusion: Establishment of human oocyte chromosome preparation and fluorescence in situ hybridization platform for the first time in Northwest China.Age is the most important factors that affects the aneuploidy of oocytes.AFC,AMH and bFSH levels did not show correlationship with oocyte aneuploidy.The application of high dose ovulation stimulation drugs may lead to aneuploid,but this study did not show the statistically significance.More research is needed to confirm this. |
