Chickpea(Cicer aretinum L),which belongs to the genus Cier(Leguminosae),is one of the most widely planted legumes.Chickpea can be eaten and used as medicine,has two thousand years of application history in Xinjiang Uygur Medicine,and has received in the "drug standard of Ministry of health of People's Republic of China for Uygur Medicine Registration" and " Uygur traditional medicine " commonly used herbs.It is commonly used to treat bronchitis,catarrh,cholera,constipation,diarrhea,indigestion and diabetes.In addition,to prevent high blood pressure,arteriosclerosis,cholesterol,reduce blood sugar and blood lipids also have certain curative effect.Chickpea is a high nutrition edible legume,sweet natured,has the function of Buzhongyiqi,Wenshen impotence.Domestic and foreign scholars have conducted in-depth research on the protein,amino acid,fatty acid,polysaccharide,trace elements and other nutrients,and the chemical constituents of flavonoids,saponins,terpenes were researched systematically.Triterpenoid saponins as one of the main active components in chickpea plays an important role in many pharmacological activities,as for its potential of pharmaceutical value,this paper carries out a systematic research on the chemical constituents of triterpenoid saponins in chickpea.The total saponin extract from chickpea has a certain degree of hypoglycemic effect,but the separation and identification of specific active compounds have not been reported.In order to make full use of chickpea resources,this study aims to separate the active saponin monomers,which can provide references for the further study of triterpenoid saponins from chickpea.In this experiment,D101 macroporous resin,ODS column chromatography and preparative HPLC were used and 11 triterpenoid saponin compounds were isolated.With the comprehensive application of HR-EI-MS,1H-NMR,13C-NMR,HSQC,HMBC,NOESY,the physical and chemical properties,structures of compounds were identified as soyasapogenol B 3-O-?-D-galactopyranosyl-(1?2)-?-L-rhamnopyranosyl(Chickpea saponin B1)(1);soyasapogenol B 3-O-?-D-galactopyranosyl(Chickpea saponin B2)(2);soyasapogenol B 3-O-?-D-glucuronic acid(3);soyasapogenol B 3-O-?-L-rhamnopyranosyl-(1?2)-?-Dgalactopyranosyl-(1?2)-?-D-glucuronopyranosyl(soybean saponin Bb)(4);soyasapogenol B 3-O-?-L-rhamnopyranosyl-(1?2)-?-D-galactopyranosyl-(1?2)-?-D-glucuronopyranosyl methyl ester(soybean saponin Bb methyl ester)(5);soyasapogenol B 3-O-?-D-galactopyranosyl-(1?2)-?-D-glucuronopyranosyl(soybean saponin Bb')(6);soyasapogenol B 3-O-?-D-galactopyranosyl-(1?2)-?-D-glucuronopyranosyl methyl ester(soybean saponin Bb' methyl ester)(7);complogenin 3-O-?-D-galactopyranosyl-(1?2)-?-L-rhamnopyranosyl(Chickpea saponin B4)(8);complogenin 3-O-?-L-rhamnopyranosyl-(1?2)-?-D-galactopyranosyl-(1?2)-?-D-glucuronopyranosyl(9);soyasapogenol E 3-O-?-L-rhamnopyranosyl-(1?2)-?-Dgalactopyranosyl-(1?2)-?-D-glucuronopyranosyl(soybean saponin Be)(10);yunganogenin D 3-O-?-D-galactopyranosyl-(1?2)-?-L-rhamnopyranosyl(Chickpea saponin B3)(11),respectively.Among them,compound 1,2,8,11 were new compounds.Study on triterpenoid saponin compounds of chickpea provides the theoretical support for the development and utilization of chickpea,and provides a new idea for the hypoglycemic effect of total saponins of chickpea.
