| Objective: Adopt irradiation of SD rat gastric with single high dose electron beam,simulated gastric intraoperative radiotherapy(IORT).To observe the injury and repair of the rat gastric after irradiation for theoretical of animal experiment for high dose IORT.Method: Thirty-eight healthy male eight weeks old SD rats were randomly divided into eight rats in normal groups(A group),thirty rats in experimental group(group B).Cooperate patent Accurate Irradiation Experiment Table for Small Animal Radiation with the thickness of 1cm electronic line bracket homemade 3×3cm2irradiation lead single 6Me V electronic line 20 Gy accuracy irradiation of group B rat gastric.After irradiation observe the general condition of rats.dietary amount and water intake and weight gain pattern and pathological changes of gastric mucosa.The average body weight and food intake data were mean ± standard.deviation.Respectively use SPSS17.0 software analysis two sets of data.Results: Single high dose electron beam 20 Gy irradiated rat gastric two weeks,group B rats activity reduction.Death rate 10%.Group B rats body hair in Irradiation region disappearing,after irradiation.The amount of diet decreased within eight days after irradiation compared with group A(P <0.05).Group B body weight decreased irradiated first week,the weight began to gain irradiated two weeks.The weight gain fastest in the irradiated from fourth week to sixth week.irradiated sixth week the amount of diet and body of two group weight no difference(P>0.05).Obvious group B rat gastric haven’t perforation and necrosis.The bright red color gastric tissue and peripheral tissue with fibrous adhesion heaviest in irradiated fourteenth days,lightest and gastric tissue become grayish yellow in irradiated fifty-sixth days.HE staining in microscopically obvious: gastric mucosal erosion,submucosal edema.In irradiated twenty-eighth days obvious submucosal edema was observed under light microscope,fibrillation,glandular atrophy,partial intestinal metaplasia.At irradiated fifty-sixth days,the gastric glands atrophy,inflammatory exudates.Group A intact gastric tissue,bright red color,cells arranged neatly,no glandular atrophy,no fibrosis and inflammatory cell infiltration,the submucosa muscularis and serosa were normal.Conclusion: SD rat gastric after 6Me V electronic line 20 Gy,the gastric without have perforation and other serious complications.Acute gastrointestinal reaction heaviest and great impact on weight and diet at rat gastric irradiated two weeks.the better time of protective treat radioactivity mucosa injury preferably opportunity.At irradiated eighth weeks complete the formation of fibrous repair.Shown in the irradiation of energy and dose can be further increased to explore the rat stomach by irradiation can withstand higher doses. |
