| Objectives1.To explore the association of DCDC2 gene polymorphisms with developmental dyslexia in Xinjiang Uyghur children.2.To explore the relationship between interactions of DCDC2 gene polymorphisms and environment and the susceptibility of developmental dyslexia in Xinjiang Uyghur children.Methods1.We selected 196 Uyghur dyslexic children and 196 controls based on the same descent,grade,sex and age.DNA was isolated from oral mucosal cell samples.Then,we used a custom-by-design 48-Plex SNPscan? Kit to genotype fourteen single nucleotide polymorphisms(SNP)of the gene DCDC2.We compared the allele and genotype frequencies of the SNPs between the two groups by means of the chi-square test.In addition to single-locus analysis,a systematic exploration of possible haplotype effects was performed using Haploview4.2 software.2.Environmental influential factors of developmental dyslexia in Xinjiang Uyghur children were preliminarily screened。Then multifactor dimensionality reduction(MDR)was used to detect the relationship between gene-environment interaction of DCDC2 and the susceptibility of developmental dyslexia.Model with the highest testing accuracy,the significant testing result and the highest CV consistency was regarded as the best.Results1.As a result,with regard to the allele frequency of these polymorphisms,three SNPs(rs807724,rs2274305 and rs4599626)displayed association with dyslexia(P<0.05),rs6456593 and rs6922023 were significantly associated with dyslexia under the dominant model and in the heterozygous genotype(P<0.05),rs9467075 and rs2274305 displayed significant associations with dyslexia under the dominant model(P<0.05).We additionally discovered that T-G-C-T of four-marker haplotype(rs9295619-rs807701-rs807724-rs2274305)and T-A of two-marker haplotype(rs3765502-1087266)were significantly higher in the DD group than in the control group(P<0.05).2.MDR analysis revealed that the five-factor combination(the condition that children finish the homework-the condition that children take part in the excellent class-family economic income-rs6456593-rs2274305)was the best gene-environment interaction model of DCDC2 in development dyslexia(testing accuracy is 0.6166,CV consistency is 10/10,P = 0.012).Conclusions1.Gene polymorphisms of DCDC2(rs6456593,rs9467075,rs807724,rs2274305,rs4599626,rs6922023)might be associated with the onset of developmental dyslexia in Xinjiang Uyghur children.2.Interactions between DCDC2 gene polymorphisms(rs6456593,rs2274305)and certain environmental factors(the condition that children finish the homework,the condition that children take part in the excellent class and family economic income)may be associated with the initiation of developmental dyslexia. |
