Background: Establish the rat model of heatstroke in dry heat environment with the pretreatment of curcumin to investigate the protective effect of curcumin on renal injury and its possible mechanism in dry heat environment.Methods: 200 male SD rats were randomly divided into three groups: normal temperature group(0 min group),the 50 min group,the 100 min group,and the 150 min group.Each group was divided into five subgroups(n=10): saline group,CMCNa group,curcumin 50mg/kg group(Cur 50mg/kg),curcumin 100mg/kg group(Cur 100mg/kg),curcumin 200mg/kg group(Cur 200mg/kg).The saline group was given 0.9% normal saline,and the CMCNa group was given 0.5% CMCNa solution.After the successful establishment of dry-heat environment rat model,the rats were anaesthetized and sacrificed at 50 min,100min,150 min and collected the blood,urine and renal tissue for further analysis.Results:(1)In the 50 min,100min and 150 min group,the content of urea,creatinine in serum and KIM-1,NGAL in urine of rats pretreated with different concentrations of curcumin were decreased at the same time compared to the saline group(NS)and sodium carboxymethyl cellulose(CMCNa)(P<0.05).(2)In the heat-dry environment,pretreatment of different concentrations of curcumin could decrease the content of MDA in renal tissue and improve the activity of CAT and SOD,the difference was statistically significant(P<0.05).(3)At the same time point in dry heat environment,the level of IL-1β,IL-6 and TNF-α of renal tissue in the curcumin pretreatment group were lower than the saline(NS)group and CMCNa group,the difference was statistically significant(P<0.05).(4)In the 150 min group,pretreatment with different concentrations of curcumin could significantly improve the pathological change of renal tissue.(5)TUNEL staining of paraffin sections of rat kidney tissue showed that curcumin pretreatment could reduce the apoptosis rate of renal cells in 150 min group,the difference was statistically significant(P<0.05).(6)In the 150 min group,curcumin pretreatment could reduce the inflammatory reaction and the expression of apoptosis related proteins in renal tissue,and the difference was statistically significant(P<0.05).Conclusion:(1)In the dry-heat environment,pretreatment of different concentrations of curcumin could reduce renal injury at different degrees.(2)Curcumin alleviated the injury of rat kidney in dry heat environment may through inhibiting inflammation and oxidative stress,cell apoptosis pathway.
