Objective:To establish the model of rat hepatic oval cell(HOC)proliferation in hepatitis microenvironment for observing the stem cell characteristics of HOC.Methods:55 male SD rats were randomly divided into the normal contrast group,chronic hepatitis group,hepatic inflammation + 2-AAF model group and 2-AAF/PH model group.Hepatitis was induced in SD rat by hypodermic Carbon tetrachloride and alcohol-containing beverage.Proliferation of HOC was induced in SD rat with hepatitis by simultaneous intragastrical administration of 2-acetaminofluorence(2-AAF)and in normal SD rat by liver 2/3 excision and postoperative intragastrical of 2-AAF respectively.The liver histology change was observed by HE staining and VG Collagen staining,and the expression of EPCAM and Thy-1 were observed by immunohistochemistry.Results:Lots of HOC were found in liver tissue of SD rat with hepatitis and gavage of 2-AAF,especially at 8th week,also in rat with liver 2/3 excision and postoperative gavage of 2-AAF,but fewer HOC were found in rat with hepatitis.HOC overexpressed Thy-1 in hepatitis microenviroment.Conclusions:Proliferation of HOC was successful induced in SD rat with hepatitis by simultaneous intragastrical administration of 2-AAF.Hepatitis microenviroment is one of the reasons that HOC tends to show characteristics of hepatocarcinoma stem cell.
