Preparation Of Centella Asiatica Compound Myogenic Gel And Study On Pharmacodynamics Of Anti Diabetic Cutaneous Ulcers

Objective:To provide a reasonable basis for further study of CACMG,we use the polymer of hydroxyethyl cellulose as matrix,combined with asiaticosides and NO,developed a new topical gel that can promote wound healing,At the same time,to establish comprehensive research and evaluation of gel,We start with the preparation process,quality standard,skin toxicity,reproductive toxicity and pharmacodynamics of anti diabetic cutaneous ulcers research.Methods:1.The study on the preparation technology and transdermal permeability of CACMGThrough the appearance,spreadability,uniformity and stability as the standard for evaluation of sensory index and experimental indexes,according to cumulative drug release rate per unit area through using the intact skin and exfoliating skin in vitro transdermal absorption,and choose the best prescription.2.The study on the quality of CACMG1)Investigation on the appearance and viscosity of gel.2)Identify of asiaticoside and madecassoside by thin layer chromatography and determination of asiaticosides in gel by HPLC.3)Investigation on stability of preparation.3.The study on the skin toxicity and reproductive toxicity of CACMG.Skin toxicity was studied by acute toxicity,skin irritation and skin sensitivity test.The animal reproductive organs were weighed and calculated the organ index,HE staining was performed,to study its reproductive toxicity.4.The Study on pharmacodynamics of CACMG anti diabetic cutaneous ulcers.Preparation of full-thickness skin defect model of normal and diabetic rats,acoording to detection of weight,blood sugar and water intake,the vitro bacterial counts,the wound healing rate,histological examination,HE staining of skin,discussing the effect of CACMG promote wound healing.Results:1.The study on the preparation technology and transdermal permeability of CACMGThrough the sensory index,experimental index and percutaneous permeability,ultimately determine the prescription that gel 1 form by hydroxyethyl cellulose 2%,8%asiaticosides,8% ascorbic acid,5% glycerol,1% azone,gel 2 form by 2% hydroxyethyl cellulose,2% nitrite,5% glycerol,1% azone.According to the permeability of skin,it was found that the effect of gel on the release rate of Exfoliating skin was greater than intact skin.2.The study on the quality standard of CACMG The gel 1 was brown yellow,the gel was colorless and transparent semi-solid gel,the viscosity was between 10-20 Pa.s,The specificity of TLC and HPLC was strong,and the negative control was not interfered.The gel has good stability at low temperature3.The study on the skin toxicity and reproductive toxicity of CACMG.CACMG had no obvious skin toxicity and no effect on reproductive organs(P>0.05).4.The study on pharmacodynamics of CACMG anti diabetic cutaneous ulcers.Successful preparation of diabetic cutaneous ulcers model.The results showed that the number of bacteria in gel group was less than model group(P<0.05).4% CACMG in the ulcer wound healing effect is the best.Conclusion:CACMG preparation method is feasible,quality easy control.Gel should be stored in4~8 ? environment,and no obvious skin toxicity and reproductive toxicity,can promote ulcer healing,to provide the basis for the further study of the preparation.
