Objective We attempt to investigate the protective effects of Xuebijing injection on lung injury in septic rats and the underlying molecular mechanisms.Methods In this work,a total of 100 Sprague-Dawley healthy adult rats(weighing 250 ± 25g)were randomly divided into sham operation group(NS group,n=25),sepsis group(CLP group,n=25),Xuebijing treated in 3 hours after CLP group(Xuebijing 3h group,n=25),Xuebijing treated in 12 hours after CLP group(Xuebijing 12 h group,n=25).Eight rats were randomly sampled in each group for survival analysis.Rats in each group were sacrificed at 6,15 and 24 h after operation,the blood and lung tissues of rats were collected.Wet-to-dry lung weight ratio and pathologic changes of lungs were measured to evaluate pathological scores of lungs.Then,serum TNF-α and IL-6 concentration were exa分钟 ed in lung blood.The expression of cytoskeletal protein Claudin4 in lung tissues was surveyed using immunohistochemistry technology.Finally,the proportion of T helper cell type 1(Th1)cells in peripheral blood cells were analyzed by flow cytometry.Results The survival rates of NS group,CLP group,Xuebijing 3 h group and Xuebijing 12 h group in 72 h after operation were 8/8,0,1/8 and 0.The survival rate of A group rats Compared with CLP group,the survival rate of A group rats was significantly increased(P<0.01)with pulmonary edema index.The detection of index of NS group after the operation indexes are all basically unchanged.The pathological scores representing lung injury were significantly reduced(P<0.05).The pulmonary edema indexes of CLP group,Xuebijing 3 h group and Xuebijing 12 h group at 15 and 24 h after operation,were significantly higher than that of NS group at the same time points(P <0.01).Pathological scores,levels of TNF-α,IL-6 of CLP group,Xuebijing 3 h group and Xuebijing 12 h group at 6 h,15 h and 24 h after operation were significantly higher than that of NS group at the same time points(P <0.01).The indexes of CLP group and Xuebijing 12 h group at each time point were not statistically significant(all P >0.05).Content of Claudin4 was significantly decreased(P<0.01).There was no statistic difference of above indexes between Xuebijing 12 h group and CLP group.Flow cytometry results revealed the proportion of Th1 cells in peripheral blood cells was increased gradually.Conclusion The Use of Xuebijing injection in the early stage could efficiently repress the inflammation of septic rats,reduce lung injury and improve the survival rates of rats.The main mechanism of Xuebijing injection might be regulating Th1 cells,delaying inflammation to strive for more therapy time. |
