Assisted reproduction techniques(ART),such as in vitro fertilization(IVF)and intracytoplasmic sperm injection(ICSI),can help sub fertile couples to create a baby.It is necessary to induce multiple follicles,which is achieved by follicle stimulating hormone(FSH)injections.At present,there are some limitations in clinical application of FSH preparation,such as short half-life,multiple injections,poor compliance,and so on.The research of FSH sustained-release preparation is of great significance.FSH sustained-release preparation based on biodegradation and biological dissolution system is a long-acting preparation of FSH,which leads to its convenient use,good efficacy and high safety.Nowadays,the research about FSH preparations mainly focuses on the preparation of long-acting FSH based on recombinant gene technology,but ignoring the long-term preparation of FSH based on the theory of polymer chemistry.Therefore,the research prospect of FSH sustained-release preparation is very broadly;its theory and application research in the clinical treatment of infertility and animal husbandry development has far-reaching significance!Hyaluronic acid(Hyaluronan,HA)derivative,sodium hyaluronate has become commonly used in drug delivery carrier in clinic.In this experiment,a FSH gel containing sodium hyaluronate was prepared.And the bioactivity of the gel was measured by ovarian weight gain method.The concentration of sodium hyaluronate was preliminarily screened.Then we research the histological of ovary and uterus of mice.KM mice were used to study the effect of superovulation.In order to investigate ovary development mechanism of mouse and the superovulation effect on ovarian development and follicular maturation.The expression of related growth factors(EGF and IGF-Ⅰ)receptor and gonadotropin(FSH and LH)receptor in ovarian development mice were studied,in order to grasp the mechanism for mature ovarian and follicular development.The above studies show that the FSH injection containing hyaluronic acid has a high biological activity,and a single injection can achieve the effect of three doses of FSH injection on ovarian weight gain.And its superovulation effect was significantly better than the natural ovulation and commercial over the row of drugs.Experiments show that the HA concentration of 5 mg/mL,FSH dose of 5IU FSH long-acting preparation of the superovulation effect is the best.This study provides a new idea for the preparation of FSH long-acting preparations for low-side effect,low-cost,good-efficacy and large-scale production applications,and has carried on the related mechanism research to elucidate the mechanism of FSH,and laid the foundation of pharmacology and medicine research.
