Objective To explore the main environmental risk factors of atherosclerotic coronary heart disease and compare the expression of NR027032,NR047116and NR104181 in atherosclerotic coronary heart disease and control group(human peripheral blood leukocytes and cultured human coronary artery endothelial cells).Analyze the association between long non-coding RNA and atherosclerotic coronary heart disease after the adjustment of common environmental risk factors and provide the clues for epigenetic molecular mechanism of atherosclerotic coronary heart disease.Method A hospital-based case-control study was conducted.A total of 516 atherosclerotic coronary heart disease patients and 516 healthy volunteers without any evidence of cardiovascular diseases were assessed from March 2015 to September 2016 at the Fujian Medical University First Affiliated Hospital,Fujian People’s Hospital and Fujian Medical University Union Hospital,China.Questionnaire survey,health examination and molecular biology laboratory testing were conducted.The contents of the questionnaire includes general demographic characteristics,smoking history,drinking and other personal life behavior factors,cardiovascular disease family history,social and psychological factors.Health examination includes the BMI index,waist and hip ratio and cholesterol levels and other blood biochemical indicators.Quantitative real-time PCR was used to profile the expression levels of the NR027032,NR034083,NR104181 that previously demonstrated to be associated with atherosclerotic coronary heart disease.All patients were fully informed of the study procedures before they provided consent.Multiple factor logistic regression was used to analyze the influence of environmental factors and genetic factors on the prevalence of atherosclerotic coronary heart disease.Result(1)Multivariate logistic regression analysis showed that overweight or obesity,type A personality,alcohol consumption and anxiety were the major environmental risk factors for atherosclerotic coronary heart disease.(2)The expression levels of NR027032,NR047116 and NR104181 in case group were lower than those in the control group(P<0.05).(3)Multivariate logistic regression analysis showedthat the low expression of NR027032,NR047116 and NR104181 was an independent risk factor for atherosclerotic coronary heart disease after the adjustment of the common environmental risk factors.(4)The expression levels of NR027032,NR047116 and NR104181 in the human coronary artery endothelial cell group(case group)were significantly higher than those in the control group(P<0.05).Conclusion Atherosclerotic coronary heart disease is a complex disease co-regulation of environmental and genetic factors.NR027032,NR047116 and NR104181 that associated with endothelial cells may be related to the atherosclerotic coronary heart disease and the change of its expression may be involved in the development of atherosclerotic coronary heart disease.Therefore,the environment-psychological-genetic medicine model should be considered fully during the prevention and treatment of atherosclerotic coronary heart disease as to prevent the occurrence and development of atherosclerotic coronary heart disease more effectively.
