Objective:Cerebral hemorrhage of rat hippocampus by autologous arterial blood reinjection as test object,detection the VEGF and cox-2 expression change through the cerebral hemorrhage within the brain tissue,research the protective effect of paeonol in damaged brain tissue at the time of the cerebral hemorrhage,and effects of Paeonol Changes of VEGF and COX-2.Method: The model of intracerebral hemorrhage was used to prepare cerebral hemorrhagic rat model.paeonol was used as therapeutic drug.The animal model of cerebral hemorrhage was treated by immunohistochemistry,Western-boltting and Elisa method.The expression of VEGF and COX-2 protein expression changes were detected.The expression of VEGF and COX-2 mRNA was detected by RT-PCR.The experimental data were analyzed by SPSS11.0 statistical software.Results:(1)The expression of VEGF and mRNA in the hippocampus of the normal group was significantly higher than that in the normal group.The expression of VEGF and mRNA in the hippocampus of the model group was significantly higher than that in the control group Higher than the model group and normal group,P<0.05;(2)The expression of COX-2 and the expression of COX-2 in the hippocampus of the normal group were significantly higher than those in the normal group,P<0.05.After treatment with paeonol,COX-2 and its mRNA expression was significantly lower than that of the model group,with significant difference(P <0.05).Conclusion:(1)The expression of VEGF in the brain tissue of the hippocampus of the intracerebral hemorrhage rats was significantly increased,the neuron resistance was increased and the microcirculation of the hemorrhagic area was reconstructed.(2)COX-2 was highly expressed in brain tissue of injured area,and the injury of nerve cells was increased.(3)Paeonol can increase the expression of VEGF,while reducing the expression of Cox-2,play a protective effect on cerebral hemorrhage neurons. |
