| Objective1.To clone human skin AQP3 gene,in order to lay the foundation for the follow-up experiment of human skin AQP3 study.2.To be ready for the preliminary experiment for a simple accurate tool products which can replace the AQP3 function,and to carry out gene cloning experiments according to the overall experimental roadmap.MethodsCloning human skin AQP3 gene complete sequence by PCR and TA cloning technique;using a variety of bioinformatics online software to analyze the nucleotide sequence of AQP3 gene and its deduced amino acid coding sequence.ResultsThrough the AQP3 gene PCR and TA cloning technology of human skin,the target product of 879 bp was obtained successfully,and the AQP3 gene of human skin was obtained,which encoded 292 amino acids;Experimental obtained positive clones plasmid DNA sequencing results in genebank sequence similarity search,find five species of Aquaporin3 amino acid sequence,their registration number respectively is: Human NM004925,Mouse NM016689,Westem clawed frog CR855446,Asiatic toad FJ605480,Dark-spotted frog FJ595953;Experimental study on the amino acid sequence about human AQP3 is similar to the result in the GeneBank.ConclusionBased on the human skin tissue AQP3 gene PCR and TA cloning technology,successfully obtained the full sequence of AQP3 gene in human skin tissue,can be used as an effective tool for post research;The positive clone plasmid sequencing results in GeneBank sequence similarity search results,and to register the gene products are consistent,suggesting that the subsequent use of the value of income. |
