Background: Endometrial carcinoma is a common malignant tumor of the female reproductive system.It accounts for about from 20% to 30% of female malignant tumor,a serious threat to women's health,life and quality of life.The disease is a group of epithelium in the endometrium of malignant tumor,the most common pathological type is endometrioid adenocarcinoma.In recent years,the incidence rate and mortality have increased gradually,and more tend to be younger.The specific course of formation of the disease is not clear,it may be related to cytokine,oncogene,tumor suppressor gene,a variety of factors related to the interaction between internal and external environment.mi R-145 is a kind of endogenous single stranded RNA molecules encoding,expression of genes regulated at the transcriptional level,participating in various biological behavior,influencing the occurrence and development of tumor,playing the role of tumor suppressor gene.IL-10 is a kind of biological activity with strong immunosuppressive factors,promoting the occurrence of tumors.Objective: To research the discrepancy expression of mi R-145 and IL-10 in the human endometrioid adenocarcinoma of various differentiation grades,and the correlation of mi R-145 and IL-10 expression.Comprehending the variation and significance of mi R-145 and IL-10 in human endometrioid adenocarcinoma.Method: Collecting 65 patients who underwent surgery because of endometrioid adenocarcinoma in the Affiliated Hospital of Qingdao University during 2011.6-2015.8,selecting samples of endometrioid adenocarcinoma tissue as experimental group,in which there are 21 well differentiated samples,27 moderately differentiated samples,17 poorly differentiated samples.Selecting 23 endometrial hyperplasias tissue after diagnostic uterine curettage as control group.Using real-time quantitative PCR to investigate the expression of mi R-145 and IL-10,and analyse the discrepancy expression and correlation of mi R-145 and IL-10 in endometrioid adenocarcinoma.Results:(1)Compared to endometrial hyperplasias tissue,the expression of mi R-145 is significantly downregulated in endometrioid adenocarcinoma tissue(P<0.05);(2)The expression of mi R-145 is decreased as the the degree of differentiation reduced,mi R-145 is positively correlated to the degree of differentiation in endometrioid adenocarcinoma tissue(P<0.05);(3)Compared to endometrial hyperplasias tissue,the expression of IL-10 is significantly upregulated in endometrioid adenocarcinoma tissue(P<0.01);(4)The expression of IL-10 is increased as the the degree of differentiation reduced,IL-10 is negatively correlated to the degree of differentiation in endometrioid adenocarcinoma tissue(P<0.01);(5)The expression of mi R-145 is negatively correlated to the expression of IL-10 in endometrioid adenocarcinoma tissue(P<0.05).Conclusions: mi R-145 is downregulated,IL-10 is upregulated in endometrioid adenocarcinoma tissue,the expression of mi R-145 is negatively correlated to the expression of IL-10.The decreased expression of mi R-145 and increased expression of IL-10 may be related to the occurrence and development of endometrioid adenocarcinoma.The body may upregulate the expression of mi R-145,downregulating the expression of IL-10 or mi R-145 through the up regulation of IL-10 through a certain mechanism to inhibite of endometrial cell lesions.It is speculated that the abnormal expression of mi R-145 and IL-10 may play a role in the occurrence and development of endometrioid adenocarcinoma.
