Anoectochilus(Anoectochilu roxburghii(Wall.)Lindl)is an orchid Anoectochilus plant and the whole plant is of medicinal use.Anoectochilus is a rare medicinal herb and called the "manual","gold grass" and "bird ginseng" by folks.Anoectochilus is generally produced in Taiwan,Fujian,Guangdong,Yunnan and other places.Because of its multiple functions,it is widely used in Fujian,Taiwan and other countries for the treatment of tumors,diabetes,hepatitis,nephritis and so on.Volatile oil extracted from Anoectochilus is composed of a variety of ingredients,including terpenoids compounds,the oxygenated derivatives of aromatic compounds(alcohols,aldehydes,ketones,phenols,ethers,lactone),nitrogen and sulfur compounds.In order to verify the anoectochilusroxburghii pharmacological effects of volatile oil,we optimized the extraction process of volatile oil,identified volatile oil components,and tested the proliferative ability spleen lymphocytes and related inflammatory factors level from mice injected with cyclophosphamide,paving the way for the immune regulation mechanism of n Anoectochilus volatile oil clinical medication.Chapters 1.Optimization of extraction conditions for volatile oil from Anoectochilus roxburghii by microwave-assisted extraction method: the material liquid ratio,extraction temperature and extraction time are optimized by single factor method,the Box-Behnken response surface method design 3 factors and 3 levels of test results: the ratio of solid to liquid 1:4(g·m L-1),extraction temperature of 60℃,microwave extraction time 20 min,extraction of volatile anoectochilusroxburghii the rate of oil was 0.0617% are determined.Chapters 2.Qualitative analysis of volatile oil of Anoectochilus.GC-MS method is adopted for qualitative analysis,and the differences of volatile constituents from different Anoectochilus samples are also compared.Results show palmitic acid are the main components,followed by 2,6--butylated hydroxytoluene,oleic acid amide,linoleic acid,linolenic acid;Other ingredients include alkanes,fatty acids,amides,esters,ketones,etc.The proportion of amide in the tissue culture Anoectochilus Roxburghii was about 12%,which was about 4 times as much as that in the artificial cultivation samples.Chapters 3.The immune effects of volatile oil of Anoectochilus roxburghii in vitro.Different concentrations of Anoectochilus volatile oil were administered into normal mice and mice injected with cyclophosphamide.Through the detection of splenic lymphocyte proliferation ability,the phagocytic activity of peritoneal macrophages and the erpession level of IL-2,IL-4 and IFN-γ,we found out that a relatively low concentration of Anoectochilus roxburghii enhance immune function in mice and higher concentration restrain immune function.
