Fosfomycin((-)FOM)is a small molecule polar antibiotic.In 1969,(-)FOM was found in the Streptomyces fradicle fermentation broth.Its chemical structure is simple,but has unique antibacterial mechanism and broad-spectrum antibiotics.Today,(-)FOM has achieved by chemical industry synthesis.(+)FOM is enantiomer of(-)FOM,produced as a by-product during the large-scale industrial production of the(-)FOM chemical industry by nearly 1: 1proportion.(+)FOM emissions are mainly divided into pharmaceutical wastewater discharge and solid stacking.Because the(+)FOM concentration within the pharmaceutical wastewater is low and the composition of the wastewater is complex,it is more suitable for its harmless degradation,in order to achieve the environmental remediation.The amount of solid(+)FOM in the warehouse is large and the purity is high.It is more suitable to convert its chiral into(-)FOM,turning waste into treasure and to improve production efficiency.Based on the above two different application purposes,this paper constructs a new screening method of(+)FOM biotransformation bacteria.The model comprises of two parts,TLC-OD570 method and ?-(+)-PEA precipitation method,which was suitable for the screening of(+)FOM degrading bacteria and(+)FOM chiral transformation bacteria.At the same time,the two methods were optimized,the accuracy was measured,the repeatability was measured,the stability was measured,and the standard curve was drawn.In this paper,samples were collected from different environments,and 476 strains of(+)FOM tolerant strains were screened as the source of screening for(+)FOM biotransformation strains.Based on the tolerant strain and the constructed detection method,20 strains of(+)FOM degradation and 1 strains of(+)FOM chiral conversion were screened.The mutant UV-35 was obtained by UV mutagenesis of strain A68-32 with highly degrading(+)FOM.The tolerant to(+)FOM was improved by 1.5 times and the degradation efficiency was increased by 26.1% The degradation rate of 1.0%(+)FOM was 87.9%.In1.0%(+)FOM culture medium after 7 days of fermentation,the strain C42 of(+)FOM chiral conversion activity conversion rate of 1.6%.At the same time,the above 20 strains of(+)FOM degrading bacteria were phylogenetic analysis and found that they belong to 10 genera:Bacillus,Brevibacterium,Brevundimonas,Cellulosimicrobium,Corynebacterium,Leucobacter,Ochrobactrum,Pseudochrobactrum,Psychrobacter and Tsukamurella,which provided a reference for the further study and utilization of the strain.
