Expression And Clinical Significance Of LncRNA UCA1 In Serum Of Healthy People,Chronic Hepatitis B Patients Or HCC Patients

Objective:To investigate the expression level of Long non-coding RNA UCA1 in serum of healthy people,patients with chronic hepatitis B and HCC,and to explore the clinical significance of LncRNA UCA1 in chronic hepatitis B and HCC.Method:30 cases were enrolled into healthy control,chronic hepatitis B and HCC,respectively.Fasting venous blood was collected and centrifuged to extract total RNA from serum.RT-PCR was used to detect the expression level of LncRNA UCA1 based on cDNA obtained by reverse transcription.Moreover,the relationship between IncRNA UCA1 and clinical characteristics of chronic hepatitis B virus as well as HCC was analyzed to demonstrate the diagnostic value of LncRNA UCA1 in patients with chronic hepatitis B and HCC.Result:compared with the healthy group,expression of UCA1 was significantly increased in the chronic hepatitis B and HCC patients(P<0.001),and was higher in the chronic hepatitis B patients than in HCC patients(P<0.05).Among the HCC patients,UCA1 expression was higher in those with more advanced TNM staging than those with earlier staging(P=0.023),higher in those with larger tumor(diameter>5cm)than in those with smaller tumor(diameter≤5cm)(P=0.005),and higher in those with poorly differentiated tumors than in those with moderately and highly differentiated tumors(P<0.001),and higher in those with lymph node metastasis than those without lymph node metastasis(P=0.018).A ROC curve of UCA1 was plotted for diagnosis of chronic hepatitis B and HCC basing on UCA1 level,and a sensitivity of 84.2%and 75%,a specificity of 72.7%and 80.0%could be achieved when the cut-off value of relative UCA1 expression was set 175.8 and 54.6.Conclusion:LncRNA UCA1 was highly expressed in serum of HCC and chronic hepatitis B patients and was related to TNM staging,tumor size,differentiation level and lymph node metastasis of hepatocellular cancer.It might also be involved in the development of HCC,and the level of serum LncRNA UCA1 possessed potential diagnostic values for HCC and chronic hepatitis B.
